Pentetrazole Repeated Kindling Influences Hippocampal Neurogenesis in Developing Rats and Its Mechanisms

Abstract


Objective To investigate the pentylenetetrazol repeatedly ignite the role in which the development of hippocampal neurons occurred and NF-κB . 72 method 10 days after birth (P10) Wistar rats were randomly divided into PTZ ( pentylenetetrazol ) group PDTC ( pyrrolidine dithio formate ) PTZ group , saline control group . The PTZ group repeated intraperitoneal injection of pentylenetetrazol to five days seizures , preparation repeatedly ignited the model ; of PDTC PTZ group, intraperitoneal injection of PDTC first , and then repeat the process of PTZ group ; saline control group , injected intraperitoneally with normal saline . Observed in each group behavioral change , and 24 hours after the model completed , three days a month , the rats were killed , were observed in the hippocampus of NF-κB expression levels , cell morphology and cell count ( thionin staining ) , BrdU (5 - bromo - , 2 - deoxyuridine ) positive cells and mossy fiber sprouting (Timm stain ) . 1 24 hours after the model completed hippocampus NF-κB expression levels of PTZ group was significantly higher than that of the control group and of PDTC PTZ group ( P lt; 0.01 ) ; models were completed three days , hippocampus neuron counts the PTZ group CA1 , CA3, and door area neuron number of the control group showed no significant difference , granule cells of the dentate gyrus (DG) area (63.13 ± 5.44) / 0.1mm2 compared with the control group (50.25 ± 6.34) / 0.1mm2 significant increase (P lt ; 0.05 ) ; of PDTC PTZ group the CA1 , CA3 , and the door zone neural element number compared to the PTZ group significantly reduce (P lt ; 0.05 ) DG district particles cell number and the control group is a significant difference ; 3 . model Preparation 3 days after completion , and DG region BrdU -positive cell number PTZ group (175.86 ± 9.20) and of PDTC PTZ group (150.75 ± 12.97) than those in the control group (131.50 ± 13.33) a significant increase ( P lt ; 0.01 ) and PTZ group significantly with more than in the PDTC PTZ group ( P lt; 0.01 ) ; 4 month after the model completed of PDTC PTZ group and the the PTZ group have mossy fiber sprouting , no significant difference between the two groups . The control group did not mossy fiber sprouting . Conclusion 1. Pentylenetetrazol repeatedly ignite can promote the development of hippocampal neurons involved in the development of hippocampal neurons process ; 2.NF-κB ; ??3.NF-κB neuron protection .
