Effect of Fosinopril on Fractalkine Expression and CD68 Positive Cell in Renal Tissue of Diabetic Rats

Abstract


Objective: To investigate Fosinopril on the CX3C chemokine Fractalkine in diabetic rat kidney and CD68 ( macrophage marker ) positive cells in kidney protective mechanism . Methods: Male Wistar rats were randomly divided into normal control group (NC ) , the unilateral nephrectomy control group ( NE group) , diabetic model group ( DM group) and diabetes fosinopril treatment group ( DF group ) , DM group and The DF group with a single intraperitoneal streptozotocin injection diabetic rat model , six in each group . Immunohistochemistry and RT-PCR, renal tissue Fractalkine expression , and distribution of CD68-positive cells of the renal tissue was detected by immunohistochemical method , and the results were semi-quantitative analysis . Results: After eight weeks of oral treatment of DM rats glucose , 24-hour urinary albumin was significantly higher ( P lt ; 0.05 ) , glomerular capillary loop area increases , the mesangial matrix mild diffuse increased with section segment increased glomerular scattered inflammatory cell infiltration . DF rats more lesions were significantly improved ( P lt ; 0.05 ) In addition to blood sugar . Immunohistochemistry chemical method and RT-PCR, DM renal tissue enhanced expression of Fractalkine ( surface density : 3.18 ± 0.45 ; mRNA : 0.53 ± 0.11 ; P LT ; 0.05 ) , glomerular CD68-positive cells was significantly increased ( 11.67 ± 0.70 / glomerular ; P LT ; 0.05 ) , the the DF set of Fractalkine expression significantly decreased ( surface density : 0.88 ± 0.29 ; mRNA : 0.23 ± 0.10 ; P LT ; 0.05 ) , while the glomerular CD68-positive cells was also significantly decrease ( 1.11 ± 0.02 / glomerulus ; P lt ; 0.05 ) , correlation analysis showed that the glomerular CD68 positive cells and Fractalkine mRNA expression was positively correlated ( r = 0.807 , P lt ; 0.05 ) Conclusion : Fractalkine expression was significantly increased in diabetic kidney tissue , and positively correlated with glomerular CD68-positive cells expressed the chemokine play an important role in the inflammatory response of diabetic nephropathy ; the DF rats Fractalkine compared with DM group was significantly lower , table Beng Hock Xin Puli protective effect on the kidney may be associated with down fractalkine expression , thereby regulating the inflammatory state of diabetes -related inhibition of macrophage infiltration .
