Abstract


[Objective] specific a research protein kinase C (PKC) inhibitor - hypericin (hypericin) rabbit chronic ocular hypertension retinal glutamate. [Methods] 32 adult rabbits, after Intracameral a-chymotrypsin, rabbit chronic ocular hypertension model to IOP gt; 30mmHg for modeling success. 12 rabbit eyes intravitreal injection of 0.1 ml hypericin successful modeling 10 d, 12 injected with saline, and the remaining three as a negative control. Another three normal rabbit blank control. Intravitreal injection 1d, 4d, 7d, 14d retinal glutamate content was determined by high performance liquid chromatography. [Results] normal rabbit eye IOP average (19.32 ± 1.45) mmHg, mean IOP of the experimental model of chronic high intraocular pressure (34 .12 ± 2.31) mm Hg. Normal rabbit retinal glutamate (1.71 × 10-5 ± 0.23 × 10-5) mmol / g, chronic high intraocular pressure rabbit retinal glutamate (4.65 × 10-5 ± 0.12 × 10-5) mmol / g, the latter than the former was significantly higher (t = 6.318, P = 0.021). Hypericin treated retinal glutamate levels compared with the same period in the saline-treated group, the difference was statistically significant (t = 7.156, P = 0.019, t = 7.933, P = 0.016, t = 11.993, P = 0.007, t = 5.276, P = 0.034). [Conclusion] chronic ocular hypertension rabbit retinal glutamate levels were significantly increased, intravitreal injection of hypericin can significantly inhibit retinal glutamate levels rise. [Objective] To explore the impact of hypericin rabbits with chronic ocular hypertension retinal ganglion cells. [Methods] 28 adult rabbits after chamber injection of a-chymotrypsin, a rabbit chronic ocular hypertension model. After successful modeling 8 rabbit as a negative control, the rest of the rabbit eye in the stability of elevated intraocular pressure the 10d after intravitreal injection 0.1ml or saline 0.1 ml of hypericin per wk 1 times, continuous 2wk. Another four normal rabbit blank control. Finally, the application of optical microscopy and transmission electron microscopy to observe the changes in the organizational structure of the retinal ganglion cells and measure the average thickness of the inner retinal layers. [Results] saline control group and negative control group, the number of retinal ganglion cells significantly reduced severe swelling of the serious degeneration of some cells, chromatin, mitochondria, endoplasmic reticulum and other organelles, the average thickness of the inner retinal layers significantly thinner. Ganglion cell morphology hypericin treatment group compared with saline mild damage to the control group, a slight reduction in the number of ganglion cells and degeneration of some cells, chromatin, mitochondria, endoplasmic reticulum lighter. Blank control group of normal rabbit average thickness of the inner retinal layers (78.62 ± 1.87) μm, average thickness of the inner retinal layers of the negative control group (59.30 ± 2.17) μm (73.80 hypericin treatment group the average thickness of the inner retinal layers ± 5.61) μm, average thickness of the inner retinal layers of the saline control group (58.75 ± 3.12) μm. Before comparing the two, a statistically significant difference (t = 3.254, P = 0.005) compared to the latter two, also a statistically significant difference (t = 3.189, P = 0.003). [Conclusion] hypericin as a specific inhibitor of protein kinase C, has a protective effect on chronic elevated intraocular pressure in rabbit retinal ganglion cells.
