Lung cancer tumors of the development of the body's immune dysfunction and tumor immune escape is closely related to, which is an important mechanism for the generation of regulatory T cells, the tumor-specific T effector cells can not be amplified to a certain level in order to eradicate the tumor. In recent years, the study found that the forked head / winged helix transcription factor Foxp3-specific expression in regulatory T cells is closely related to its development and functional maturation, can to some extent reflect the level and functional activity of regulatory T cells. Objective To detect the expression levels of the transcription factor Foxp3 in peripheral blood and tumor tissue of patients with lung cancer, the analysis of different pathological types of regulatory T cells and lung cancer, TNM stage and lymph node metastasis, and surgical treatment of the relationship, and to investigate the regulation of lung tumor immune and its clinical significance. Method 1. Collected peripheral blood mononuclear cells (PBMC) separation and tumor-infiltrating lymphocytes (TIL) separation of peripheral blood of lung cancer patients before and after surgery and normal controls by Ficoll density gradient centrifugation isolated peripheral blood mononuclear cells were taken; fresh lung tissue and extract the tumor infiltrating lymphocytes. By RT-PCR assay for detecting lung cancer patients and normal controls peripheral Foxp3 expression levels in the tumor tissue, and to analyze the correlation. Results 1. Lung cancer patients with tumor-infiltrating lymphocytes in Foxp3mRNA relative expression levels of relative expression level was significantly higher than that of lung cancer patients and normal control peripheral blood; lung cancer patients in Foxp3mRNA relative expression level than normal controls were also significantly elevated. 2.Foxp3mRNA expression level and the degree of local infiltration of tumor size and tumor in the peripheral blood and tumor tissue of patients with different pathological types of lung cancer, small cell lung cancer patients with high expression levels, followed by adenocarcinoma, squamous cell carcinoma of the lowest. Later TNM stage, lower tissue differentiation, lymph node metastasis the positive group Foxp3mRNA the expression was significantly increased. 3. The postoperative Foxp3mRNA expression level was significantly reduced. Conclusion 1.Foxp3 CD4 CD25 regulatory T cell-mediated immune suppression and the occurrence of lung cancer development and prognosis, surgical removal of the tumor, can reduce the body's tumor-bearing environment and Treg cells, relieve the inhibition of tumor immune function, help postoperative recovery of immune function. 2. Effective tumor biological treatment, reduced the expression of Foxp3 gene, to reduce the number and function of Treg cells, will help improve the body's anti-tumor immune to improve the treatment of lung cancer.
