Objective: To observe the surgical microscope , biochemical aspects of SD rats after exposure of the retina pathological change ; explore the effect and mechanism of Chinese herb extracts puerarin injection on rat retinal light damage repair . Methods: SD rats were randomly divided into normal control group , model group , high dose group and low dose treatment group , in addition to the normal group , each group of experimental rats accept the 22000 ± 1000lux surgical microscope light source 48 continuous irradiation for 30 minutes two treatment groups for consecutive day after irradiation week later, all animals were sacrificed morphological changes observed in each group between the retinal tissue and the retina of inducible nitric oxide synthase ( iNOS ) , cytochrome c (Cyt-c ), Caspase-3 content differences . Results : ① tissue morphology , the normal group, normal rat retinal morphology , retinal thinning of the outer nuclear layer of the model group than normal cells arranged group sparse , inside and outside the festival disorganized, unclear boundaries ; retinal morphology of low - dose treatment group improved , high- dose treatment group compared with the model group, no significant improvement . ② puerarin can regulate the photoreceptor apoptosis gene expression : by inhibiting INOS to reduce the over-expression of NO in the retina , thereby reducing the toxic effects of NO on the retina , and reduce Cyt-c, Caspase-3 expression , thereby inhibiting retinal apoptosis. Conclusion: This study successfully established a model of acute retinal light damage . These results suggest that low-dose treatment group puerarin reduce retinal light damage , as well as to promote the role of the retina after light damage repair . Its mechanism of action may be to reduce the toxic effects of NO on the retina , inhibit photoreceptor apoptosis .
