Objective: To investigate the protective effect and its mechanism of puerarin injection on Acute retinal light damage . : From tissue morphology and immunohistochemistry to observe the outer nuclear layer of rat retinal tissue morphological changes and detection of apoptosis regulatory factor of TNF- α , FasL, NF - κB , Caspase - 3 . Results: (1 ) tissue morphology , model control group of retinal thinning of the outer nuclear layer cells decreased cell arrangement than the normal control group sparse disordered arrangement of inner and outer section boundaries unclear ; improved retinal morphology of low - dose treatment group , retinal thickening of the outer nuclear layer compared with the model group , the cells arranged in dense, high- dose treatment group compared with the model control group had no significant improvement . ( 2 ) light damage in the retina of the model control group TNF-α, FasL, Caspase-3 expression enhanced , NF-κB expression decreased ; retina of low - dose treatment group TNF - α, FasL , decreased expression of caspase -3 protein , NF -κB protein expression increased , and the difference was statistically significant (P <0.01 or 0.05 ) ; high dose treatment group compared with the model group , the difference was not statistically significant ( P > 0.05 ) . Conclusion: This study successfully established a model of acute retinal light damage . The experimental results suggest that puerarin injection to reduce retinal light damage , protect the function of the retina , and to promote the role of the retina after light damage repair , its mechanism of action may be related to enhanced protein expression of NF-κB , attenuated TNF -α , FasL, and Caspase- 3 expression , thereby inhibiting retinal cell apoptosis .
