Abstract

Methods: Kunming mice as experimental model of morphine withdrawal. 1 mice were randomly divided into model group, model EA group (referred to as the mode pin group), the model agonist group (referred to as the excited group), the model excited EA group (referred stimulated acupuncture group) and blank control group (referred to as blank group) of 5 groups, I excited group and stimulated acupuncture group were given intraperitoneal injection of 5-HT 1A receptor agonist 8-OH-DPAT (0.2mg/kg); 2. laboratory mice randomly divided into model group, mold acupuncture group, model antagonist group (referred to as the antagonistic group), antagonistic EA group (referred to pluck the pin group) and the control group of five groups, to the antagonistic group and pluck the needle group were intraperitoneally injected with 5-HT 1A receptor antagonist WAY100635 (0.5mg/kg). Baihui acupuncture take Sanyinjiao hole, then EA therapeutic apparatus, density wave, frequency 15-25 times / min mouse leg gently shake degrees, 1 / d, 15min / , treatment 3d. Using the elevated the plus maze observation 5-HT 1A agonists and antagonists on acupuncture anti anxiety behavior in morphine withdrawal; using immunohistochemistry combined with computer image analysis observed 5-HT morphine withdrawal 1A receptor agonists and antagonists on acupuncture anti anxiety sew expression of 5-HT in the nucleus and prefrontal cortex neurons in the mouse brain. Results: 1. Behavior: (1) model mice OT% and OE% value than the control group was significantly lower (P <0.01); mold pin group, excited group and stimulated acupuncture group OT% OE% values ??were significantly high in the model group (P <0.01). (2) antagonist mice OT% and OE% value blank group was significantly lower (P <0.01), slightly lower than the model group; the antagonistic needle mice OT% and OE% value than antagonistic group a significant increase (P < 0.05, P <0.01), and slightly higher than the mode pin group. 2.5-HT expression: (1) the model group and blank group raphe nuclei and prefrontal cortex neurons optical density of 5-HT significantly lower (P <0.01), the gray value was significantly higher (P <0.05); mold needle group, excited group and excited needle group and model group comparison seam nuclear and the prefrontal cortex nerve yuan 5-HT in optical density value was significantly higher (P <0.05 or P <0.01), the gray value significantly with lowering (P <0.05 or P <0.01) compared with the control group, no difference was found. (2) antagonist group and blank group was significantly lower (P <0.01), the gray value was significantly higher (P <0.05), compared with the raphe nucleus and prefrontal cortex neurons optical density of 5-HT changes consistent with the model group raphe nucleus and prefrontal cortex neurons 5-HT optical density value; pluck the acupuncture group compared with the antagonistic group increased trend the gray value significantly lower (P <0.05, P <0.01). Conclusion: 1, EA has the effect of anti-morphine withdrawal anxiety, and have a synergistic effect with 5-HT 1A receptor agonist, suggesting that the 5-HT 1A receptor anxiolytic effect is electric needle play an important way. 2,5-HT 1A receptor antagonist, failed to antagonize the the EA effects, suggesting that 5-HT 1A receptor, although an important way to play for the electro-acupuncture anxiolytic effects, However, the non-unique ways, yet there may be other effects link.
