Mastitis in dairy goats and dairy farms is often one of the diseases, mainly breast inflammation caused by microbial infection. In this study, the laboratory goat Staphylococcus aureus and Escherichia coli clinical mastitis milk samples isolated from milk Guanzhong dairy goats with clinical mastitis model established by the nipple tube perfusion method, ELISA method on five proinflammatory cytokine detection, mastitis Inactivated Vaccine against both pathogens. The main content and the results are as follows: 1. Guanzhong dairy goat mastitis model to establish isolated in our laboratory, dairy goat mastitis pathogens Escherichia coli and Staphylococcus aureus, according to E. coli 3 103cfu/ml Staphylococcus aureus 3 × 102 cfu / ml dose, respectively nipple tube perfusion health Guanzhong dairy goat, milk yield, mental condition was measured at different time points to observe dietary For milk appearance, general characteristics of the breast, body temperature, body cell number (LMT Act) and other indicators, and pathological examination and Pathogen. The research results show that the spirit of the dairy goat attack bacteria malaise, reduced feed intake, breast discoloration, swelling, fever, obvious tenderness and other symptoms of acute inflammation, milk samples discoloration, viscous, fever, reduced milk yield ; sharp increase in the number of somatic cells in the milk; biopsy revealed breast bubble and interstitial inflammatory cell infiltration; compared with E. coli, the more severe clinical and pathological changes caused by staphylococcus; side of milk from each experimental group attack bacteria samples isolated from E. coli and Staphylococcus aureus. Successfully established a dairy goat Escherichia coli and Staphylococcus aureus dairy goats clinical mastitis model. 2. Explore Staphylococcus aureus mastitis and E. coli mastitis and promote changes in the expression levels of inflammatory cytokines and their role in the course of the disease mechanisms. Respectively perfusion before and at different time points after perfusion acquisition of Staphylococcus aureus and Escherichia coli experimental group milk goat milk samples and serum the ELISA assay IL-1, IL-4, IL-6, IL-8, and IFN-γ expression levels, analysis of the relationship between the Staphylococcus aureus mastitis, Escherichia coli mastitis clinical and pathological features with these cytokines. The results showed that the two pathogens can lead to the increase in milk samples of five kinds of cytokine expression in mastitis, E. coli group reached the peak at 24h, and then reduce and maintain a certain amount of expression, Staphylococcus aureus group within 72h continued to rise. Serum cytokines generally tended to decrease expression levels even lower than before reperfusion Staphylococcus aureus mastitis E. coli infection mastitis trend of serum cytokines showed different laws. 3 homemade mastitis vaccine and the antibody titer after immunization for evaluation. With the formaldehyde 37 ° C overnight inactivated E. coli, Staphylococcus aureus, were prepared propolis inactivated vaccine, transfer factor inactivated vaccine without adjuvant vaccine; immune sheep serum and milk were collected at different times after inoculation lactation dairy goats, ELISA measurement of serum antibody titer for E. coli and Staphylococcus aureus. The research results show that the E. coli and Staphylococcus completely inactivated by formaldehyde; compared with the pre-immune serum and milk E. coli and Staphylococcus-specific antibody titers were significantly higher; compared with non-adjuvanted vaccine The propolis and transfer factor can significantly improve specific antibody titers in the blood and breast milk transfer factor to promote anti-? secretion best results; immune content of breast milk sIgA significantly improve. Inactivated Staphylococcus aureus and Escherichia coli after immunization can stimulate the level of specific antibody titers in the blood and breast milk of dairy goats, transfer factor on the promotion of the role of antibody titer after vaccination is better than propolis.
