Abstract


Brucellosis (Brucellosis) is a zoonotic disease caused by Gram-negative bacteria parasitic intracellular Brucella , mainly caused by livestock stillbirth , miscarriage , infertility and other symptoms , and affects not only the development of animal husbandry can also lead to huge economic losses and serious public health problem . In this study, widely used in China goat pox attenuated vaccine strains for the parental virus , build the Brucella the protective antigen omp31 and omp2b recombinant goat pox virus in order to obtain the recombinant sheep pox immunity against brucellosis virus , brucellosis , sheep pox prevention provide a theoretical basis . The primers were designed according to GenBank accession sheep species Brucella omp31 and omp2b gene sequence , respectively the PCR amplified omp31 and omp2b gene , digested , sequenced to verify its correctness . Identification the right omp31 and omp2b cloned into the the GTPV (Goat poxvirus) recombinant vector pGM - TK- P11- of GPT1 build reorganization goat pox virus transfer vector containing the the Brucella omp31 and omp2b gene . The constructed two goat pox virus the transfer vector GTPV of vaccine strains G14-STV44-55 transfected sheep testicular cells , screening, identification recombinant virus rGTPV strains and cell lysates of the recombinant virus expression OMP31 and OMP2b - vivo protein by Western blot , indirect ELISA to detect rGTPV strains of mice immunized with IgG antibody levels . The the experiments stable expression the Brucella OMP31 and OMP2b of protein recombinant goat pox virus strain by G14-STV44-55-BOMP31 and G14 - STV44 -55 - BOMP2b ; Western blot showed that the expression of recombinant goat pox virus OMP31 and OMP2b of protein immunoreactive ; rGTPV strains of mice immunized with test showed that the recombinant viral expression OMP31 and OMP2b of protein to produce antibodies having a certain degree of immunogenicity . In this study, to further carry out GTPV live vector vaccine and related basic research provides the technology platform .
