Abstract


Infectious bursal disease (IBD) is one of the important infectious diseases, prevention and control of it over the years against the poultry industry relies mainly on conventional vaccine immunization. But the application of conventional vaccine has its inevitable drawbacks: the residual live virus inactivated vaccine may lead to potential infection; various types of attenuated vaccines the bursal have different degrees of damage role; in addition to widely used attenuated vaccine is not conducive to infection quarantine and control of the disease, can lead to viral virulence and antigenic variation. To this end, the experimental study of infectious bursal disease anti-idiotypic antibody (anti-Id) vaccine, designed to overcome the drawbacks of the conventional vaccine, new vaccines to explore the development of a safe, non-polluting, its in the production practice in the future application of the theoretical basis for the control and eventual elimination of IBD and other infectious diseases immunization explore a new idea. In this study, prepared in the traditional serological methods polyclonal anti-Id. First in chicken embryo fibroblasts (CEF) the propagation of IBDV D 78 strains attenuated immune the primiparous SPF hens purified by sucrose density gradient centrifugation. Collection hyperimmune egg fat, chloroform, Na 2 SO 4 salting combined DEAE 3 2 ion exchange chromatography final purity high anti-IBDV-specific the yolk IgG (Ab1). This IgG as immunizing rabbits, rabbit antiserum obtained with anti-Id (of Ab 2 ). Some rabbit antiserum by (NH 4 ) 2 SO 4 salting, and then absorbed purified by affinity chromatography, was purified anti - id. Immunochemical identification of the anti-Id, ELISA competitive inhibition and inhibition of experimental results show that the anti-Id able to the IBDV compete with the original antigen binding Ab1 also able to inhibit the binding of Ab1 with IBDV, and with the increase in the concentration of anti-Id, competition and inhibition effects are enhanced. That this anti-Id contain the antigen \Functional identification of in vivo experiments, the anti-Id, chick embryo, and inhibition of experimental results show that the anti-Id antibody with IBDV and binding it lost and capacity, from a biological point of view to verify the anti-Id simulates the original antigen functions. Immune protection Experimental results show that the anti-Id vaccine chicken, no side effects, can protect chicks against IBDV attacks. Its rate of protection and the use of adjuvant, Freund's adjuvant (protection rate of 83.3%) was better than ordinary white oil adjuvant (61.5%), general anti-Id immune effect (protection rate of 69.5%) is weaker than conventional the IBD vaccines (80%).
