Studies on the resistance induced by β-aminobutyric acid against Phytophthora capsici in pepper plants and its resistant mechanism

Abstract


In plant disease control, research the chemical inducer induced resistance mechanism has important theoretical and practical significance. In this study, for the first time in the country as a non-protein amino acid beta-aminobutyric acid (β-aminobutyric acid, referred to as Baba) inducer spicy (sweet) pepper three varieties of \the induction. Foliar spray BABA inoculation of IRRIGATING method, the results show that, the Baba can induce pepper root and whole plant system resistance (SAR). The effect of high concentrations of the Baba (1000μg/ml) induced foliar applications, and no toxic effects on the plant. This induction effect by the the pepper growth duration, eggplant door bell pepper spraying 1000μg/ml Baba four-leaf stage induced resistance was 44.5%, the eight-leaf stage induced resistance up to about 70%, the second branches of almost completely to prevent Phytophthora infection; 3d, Baba induction treatment plants began to show a higher induced resistance, this induced resistance sustainable 20d. The experiments also showed that BABA has no effect on the of Phytophthora capsici wire growth rate and other culture traits, no inhibitory effect on Phytophthora, that BABA treatment pepper seedlings the induced resistance Baba can be used as inducer. Acid-soluble the the leaf homogenate protein electrophoresis and after vacuum filtration, low-speed centrifugation extracted the intercellular protein electrophoresis results show that BABA treatment 1d, 3 varieties are induced by a low molecular weight (about 16.6KD) intercellular specific protein; the CV98 \The increase in the amount of protein, without the generation of new proteins. Three varieties BABA treatment after the first 3d, have a molecular weight of about 10. SKD alkali-soluble intercellular accumulation of specific proteins in the molecular weight <14. Shape D region from an increase in the amount of the alkali-soluble specific protein. These specific protein molecular weight is relatively small, 10-40KD has strong stability, throw natural soluble at low pH T, mainly distributed sub-cell gap c with PR protein characteristics exactly match. Enzyme activity assay results showed that the leaves Bu 1,3 - glucanase, chitinase id BABA treatment that activity is enhanced. BABA treatment 4d range, three varieties of P-1, 3 - glucanase activity increased four times Bu chitinase activity by BU 12.5 times. Chitinase activity increased the magnitude of the eggplant door bell pepper is less than \specific protein, the more resistance the stronger. Enzyme activity assay results and protein electrophoresis showed the specific protein electrophoresis may belong Bu B 3-glucanase and chitinase stuffed them involved in the expression of induced resistance, and plays an important role. BABA induced increased peroxidase enzyme activity appeared in pepper leaves, peroxidase may also be involved BAEA induced expression of resistance. Far BABA induced resistance mechanism of only child of tomato late blight (corpse cut hs twist volumes), the present study, vacuum filtration, low-speed centrifugation method of acid-soluble and alkali-soluble protein extraction and electrophoretic analysis, combined with the PR protease activity analysis shows that, the PR protein plays an important role in the mechanism of induced resistance of the BABA, peroxidase may also participate in the expression of the resistance. These results provide a theoretical basis will induce the development and application of Hangzhou agent Baba.
