Powdery mildew is one of the main diseases in wheat production in China. Due to the promotion of varieties resistant to powdery mildew has anti-source single easily generate new physiological race and resistance to degradation, and therefore need to find a new resistance gene. Has been reported in the the wheat varieties Fuzhuang 30 there is a presence of powdery mildew in the 4D chromosome on the new anti recessive gene. Recessive resistance genes in conventional breeding performance of low frequency, but also to go through the turns multiple generations of backcrossing and selfing, spend a lot of manpower, material and require a longer time to breeding new varieties of disease-resistant . Molecular marker technology developed in recent years to some of the DNA fragments tightly linked with the recessive gene analysis of molecular markers in the indoor screening resistant plants as backcross parent, greatly improve breeding efficiency, can save more than doubled time, thus speeding up the process of breeding for disease resistance. In this study, the most commonly used simple the fast random amplified polymorphic DNA (RAPD) analysis of molecular markers of this disease resistance recessive gene Pmx, assisted selection for the future application of molecular markers in breeding for disease resistance in provide a theoretical basis. Experimental results show that: 1. Cluster segregation analysis (BSA) marker screening. \Three varieties of DNA as the template for RAPD analysis. Screened a total of 313 primer, F has six characteristics with primer and six missing feature with primers screened; screened seven primers with characteristic bands for the N and J two susceptible varieties. 2. F has characteristic bands the six primers confrontational varieties Rejuvenation 30 \ 2 generation of resistant and susceptible plants genetic separation of the statistical analysis. The results show that: the only primer UBC405 produced characteristic bands (UBC 405-628 close linkage between the recessive resistance gene, and F 2 generation exchange strains of the recombination rate of 13.6%, Mapmarker3.0 analysis UBC 405-628 genetic map distance of the recessive resistance genes with molecular markers 13cM, LOD = 7.3. 3. \The No.1 \628 F 2 -generation assisted selection can greatly accelerate the process of breeding resistance to powdery mildew, has important theoretical and practical significance for resistance to powdery mildew breeding.
