Abstract

Tobacco is one of the important cash crops. The tobacco wildfire disease has become the major diseases of tobacco production, in case of suitable climate easily cause a pandemic leaving tobacco production suffered significant losses. Foster anti-wildfire disease of tobacco varieties is the most cost-effective measures to prevent wildfires disease. Molecular marker-assisted breeding can shorten the breeding process, to avoid the cumbersome process Inoculation improve breeding efficiency. In this study, mainly includes four aspects: (1) to determine the appropriate tobacco wildfire indoor inoculation method. (2) determine the genetic law of the tobacco wildfire disease resistance gene from Nicotiana longiflora. (3) Construction and Speight 's G-140 (susceptible variety the) × Ky14 (disease-resistant varieties) F2 segregating population, RAPD analysis of tobacco wildfire disease resistance gene BSA method to find and close the wildfire disease resistance gene RAPD marker linked to. (4) the use of molecular marker-assisted selection and validation [(SpeightG-140 × Ky14) × Speight G-140] BC1 population. The main results obtained in this study are as follows. (1) Indoor inoculated tobacco Pseudomonas syringae pv acupuncture combined with needle-free injection method is ideal. The specific method is to use a syringe needle gently pierce the epidermis, the vaccination easier; reuse a syringe without needle injection of bacterial suspension control inoculum size, and after the onset of the lesion more rules, investigation lesion diameter more convenient and feasible. Tobacco wildfire disease when inoculated, under the premise that ensure the viability of the pathogens, in addition to the vaccination methods of choice must also pay attention to the control of humidity and temperature, especially the control of the humidity. The temperature is preferably controlled in the 26-30 ° C, the relative humidity should be maintained at more than 80%. The (2) Tobacco the wild species N.longiflora of the wildfire 0 Race Races immune. This study further verified from N.longiflora wildfire disease resistance is controlled by a single dominant gene, and the gene of resistance to their offspring can be stabilized, N.longiflora as tobacco wildfire disease resistance sources is still valid. Speight G-140 (3) of this study is to contain the Ky14 of the wildfire disease from N.longiflora resistance gene resistant parent to not contain the resistance gene susceptible parent, hybrid combinations. BSA method of resistance genes from N.longiflora RAPD analysis. Screened from 423 random primers of 10 bp RAPD markers tightly linked to the resistance gene S522000, the tag and the target gene genetic distance of 4.88 cM. (4) Using the tag S522000 [(Speight G-140 × Ky14) × Speight G-140] BC1
