Abstract
Using parthenMentlc Inducing line s polllna比Cthe stUdies on the Inducing haploids from different genotype female materials was carried ow.The n扯wal fertility ofear and inflorescence of the Induced h叩fold was observed.Some haPlold p枯ms were treated sep盯ately with different colchiclne concemration nd 2%DMSO in six leaf nd elongation stage to double their chfomosomes.At the same time flowering condition of the male Inflorescence was observed.The result硼some haplolds had nthers but c。uld n。1Shed,S。p。11en S[I侧fo1es8wem mMe 10 these hs帅The 方rtllity-restoring mechanism of the haplold was analyzed using cytology method.The pollen sheddinghaploid plants were selFpollinated to getdouble haploid(DH)lines. Agronomlc traits was observed,and polyacrylamlde gel electrophoresis(队GE)of Peroxldase(POD)and Esterase(EST) lsompe was done to DH lines. Although90.52%ofhaploidears were fertile,the fertileness ofthetassel were ic风 and pollen sheddl旭 ability wasdifferent betweengenotypes.The lowest shedding rate was 3.03%,while the hehest reacheu as h炒 as 50.00%.So the ability of the haploid seed-setting by sc卜pollination was resmctCd by the feftileness of眺咒l.yor haplold chromosome doubling treatments with different colchicine nd 2.0%DMSo,the best result Is the treatment which was made In six ie对st昭e with 0.5%colchlclnes concentration.It was be地r than 0.1%concentration.In elongation st昭e the rate of drug lm"red plants was high,and the change of*the haploid doubling rate was little.gll of*the tested haplolds have stalnable pollen.The叫erage frequency Is 8.0 po比ent.The highest Is 20.0 percent.The study also found that dlploid cell exited both In somatic cells and In pollen mother cells.Only lfdlplold cell was In somatic cells,It would appear In PMCs. The DH lines showed highly uniformity In all traltsthat can be measured.Some ofthem 41could be accepted for breeding pn吓Dsc ingrowing,ear size nd other%ronomlc traits.The results of the PAPAGE of POD and EST isowme showed that some bands of the DHlines was the same s the male material and some was the sme s the female material.ItIs therefore concluded that the呻henogentlc Inducing line Induced haplold Is a randomprocess.
