Abstract


MPTP can lead to the pathogenesis of PD has an indisputable fact , recent studies have shown that MPTP is located on the cell membrane monoamine oxidase B oxidized to MPP ~~ work to play . This study proposed by MPP ~ on cultured midbrain dopaminergic neurons observed the possible mechanism of MPP ~ induced PD onset and preparation MPP ~ vitro PD model may be its value . In order to observe the glial cells in which the role of cells into unsuppressed glial cells and glial cells inhibited group ; different concentrations of MPP ~ vitro midbrain dopaminergic neurons in the culture medium , and added Iloprost research whether there is a protective effect on the neurotoxicity caused by MPP ~ . Determination of [~ 3H] dopamine and [~ 3H] choline uptake and intracellular dopamine content and TH immunocytochemistry staining to observe the changes . The main results are as follows : 1 . MPP ~ vitro midbrain dopamine neurons [~ 3H] dopamine uptake significantly inhibited force ; inhibition in inhibition of glial cells was significantly stronger than inhibit glial cell group ; MPP ~ dopaminergic nerve the yuan [ 3H] choline uptake force no significant inhibition; 2. MPP ~ role in midbrain dopamine neurons can significantly reduce intracellular dopamine content reduced TH -positive cells in the cell body the the cell processes thinning retraction , also significantly reduced the number of TH -positive cells ; 3. No significant differences between the different concentrations of MPP ~ group ; 4 . Iloprost neurotoxicity caused by MPP ~ no significant protective effect ; Our results prove : MPP ~ significant inhibitory effect on cultured midbrain dopaminergic neurons function , which may be related MPP ~ induced dopaminergic nerve yuan related to the inhibition of dopamine uptake as well as a decrease in dopamine content , MPP ~ no significant inhibition of cholinergic neurons . Produced by MPP ~ PD cell model as an in vitro model of PD research .
