Abstract


Membrane chromatography is to maximize advantages to maintain the three - dimensional structure and activity of the integrity of cell membranes and membrane receptors . Steric interaction between the drug plasma membrane receptor ( hydrophobicity, charge , hydrogen bonding ) will be different by chromatography various quantitative characterization parameters reflect . Membrane chromatography without radioactive ligand mark in a natural membrane receptor active state , without artificial membranes were prepared , simple, a membrane column can be repeatedly applied, improve work efficiency . Demanding cell membrane chromatography stationary phase and column flow , pH , column temperature . The innovation of this paper is that , by the sol - gel prepared membrane carrier , cell membrane and drug interactions by capillary electrophoresis and capillary membrane chromatography separation of beta - lactam antibiotics . The paper is divided into three parts: Chapter 1: Introduction of the principle of capillary electrophoresis and application of sol - gel technique and application , bio- chromatography in pharmaceutical analysis . Chapter II: capillary electrophoresis method to study the cell membrane and beta - lactam drug interactions , discover the capillary membrane chromatography study drug and cell membrane interaction , the application of the new drug screening . The ScS experiments membrane coating amount , the buffer pH, the type of cell membrane retention factors (human red blood cells , erythrocytes , S-180 cells ) of the drug . Chapter III : the use of the capillary membrane chromatography to beta - lactam antibiotics were isolated and investigated the influence of various factors on the separation of drugs . Experimental results show that the capillary membrane chromatography can be directly used to study drug -membrane interactions , resulting chromatographic parameters on the mechanism of drug action and new drug development and screening has important reference value .
