Abstract


Objective: To observe the peripheral blood of patients with acute myocardial infarction expression of the mononuclear cells Fgl2 thrombin original enzymes and serum TNF-α, and comparison with stable angina and normal control subjects to explore Fgl2 and TNF-α in acute myocardial infarction and significance. Methods: Union Hospital inpatient department of Cardiology hospitalized patients as research subjects, including acute myocardial infarction (AMI) 29 cases of patients with unstable angina (UP) 28 cases, voluntary normal subjects (CG) 18. Authorized all objects venous blood was collected to obtain blood samples, blood samples of patients with acute myocardial infarction group acquired acute onset within 24h, blood samples of patients with unstable angina Get the day of admission, the normal control group for any time. All samples were never used thrombolytic agents and anticoagulants. Immunofluorescence, westen-blot detection method were used to detect the expression of peripheral blood samples mononuclear cells (PBMC) Fgl2 and each group of samples of human serum concentration of TNF-α were measured by ELISA. Results: 1. The immunofluorescence showed acute myocardial infarction group and Fgl2 of expression mean optical density value is significantly higher than unstable angina and the control group, the difference was statistically significant (P lt; 0.05). ELISA method results show that the human group with acute myocardial infarction in patients with TNF-α concentration was significantly higher than with stable angina and the control group (76.20 ± 5.79,64.55 ± 5.00 vs 38.54 ± 1.98,35.92 ± 2.03, P lt; 0.05), unstable angina group and control group difference was not statistically significant (38.54 ± 2.03 ± 1.98 vs35.92 P gt; 0.05). Results show 3.westen-blot the normal group Fgl2 protein concentration is very low, almost no expression. Unstable angina, a small amount of expression of acute myocardial infarction the group Fgl2 high expression. Conclusion: 1. Acute myocardial infarction in peripheral blood mononuclear cells Fgl2 expression was significantly increased, acute myocardial infarction, coronary microcirculation thrombosis predictive value of one of the acute phase proteins. 2. Acute myocardial infarction serum inflammatory factor levels of TNF-α increased, prompted the development of TNF-α-mediated acute myocardial infarction. Objective: To observe coronary ischemia and reperfusion in rat peripheral blood mononuclear cells in thrombin the original enzyme Fgl2 and serum levels of TNF-α expression, explore FGL2 and TNF-α in ischemia-reperfusion injury significance and relationship. Methods: Animal group: 56 male Sprague-Dawley rats were randomly divided into normal control group (n = 8 only), is divided into sham group (n = 24 rats), model group (n = 24 rats), model group ischemia-reperfusion one day, three days, seven days three sub-groups, each group of rats were sacrificed at the corresponding point in time, specimens were taken for the detection. ELISA method to detect serum cytokines TNF-α levels ischemia reperfusion area of ??heart tissue, Westen-blot detection of cardiac microvascular in fgl2 protein expression. Results: 1. ELISA method results in the surgical group, TNF-α concentration was significantly higher than the sham group and normal group (40.68 ± 6.73,66.36 ± 4.05,67.12 ± 3.88 vs 30.78 ± 6.24,27.36 ± 6.20, P lt; 0.05), sham group and normal group difference was not statistically significant (30.78 ± 6.24 vs27.36 ± 6.20 P gt; a 0.05) 3.westen-blot results showed that the the animal group normal group and the sham group Fgl2 protein concentration is very low, almost no expression. The surgery group Fgl2 protein highly expressed, the concentration of 3,7 days than one day. Conclusion: 1. Coronary ischemia reperfusion rat cardiac microvascular micro thrombosis, level of microvascular fgl2 protein expression was significantly increased, may be one of the mechanisms microcirculatory disturbance after ischemia-reperfusion. Ischemia-reperfusion Fgl2 expression of TNF-α concentrations have increased, and a positive correlation prompted micro thrombosis is an immune coagulation process.
