Abstract


Objective: To investigate correlations of the polymorphisms and methylation of HLA-DRB1 and HLA-DQB1 with the risk of Kazakh Esophageal carcinoma in Xinjiang and the significant of Clinic pathologicalMethods: 1.The polymorphisms of HLA-DRB1*0901、1501,DQB1*0301、0602 were typed by sequence specific primer based on polymerase chain reaction (PCR-SSP) in 200 Kazakh patients with esophageal carcinoma and 150 normal esophageal mucosa,the results confirmed by sequence analysis; 2. the HLA-DP, DQ, DR protein expression was detected by immunohistochemical SP method in 66 Kazakh patients with esophageal carcinoma and 28 adjacent of cancer tissue;3. HLA-DRB1 and DQB1 methylation status were Detected by MALDI-TOF MS spectrometer in 66 Kazakh patients with esophageal carcinoma ,20 adjacent of cancer tissue and 25 kazakh normal esophageal mucosal tissues ;Date deal with the software SPSS13.0 statistics .Results:1. The results polymorphisms of HLA-DRB1 and HLA-DQB1: (1)200 Kazakh esophageal carcinoma HLA-DRB1* 1501、0301 and HLA-DQB1*0602 allele frequencies were 0.455,0.76 and 0.69, the different has statistic significant compare with the genes of 150 normal esophageal mucosal tissues(0.232,0.52,0.554)(OR=2.78,OR=2.93,OR=1.80,P <0.05),the different was especially in 82 HPV infection Kazakh Esophageal carcinoma(OR=3.18, OR=2.37,OR=1.92 ;P <0.05);(2) HLA-DQB1 * 0602 allele frequency in 124 poorly differentiated Kazakhstan esophageal carcinoma(0.742) is higher than well-differentiated carcinoma(0.597) (P <0.05); 2.HLA HLA-DP, DQ, DR Protein expression: HLA-DP, DQ, DR positive expression rate was 36.4% in 66 Kazakh cases of esophageal carcinoma, 28 cases of group was 7.1%, the difference was statistically significant; 3. The result methylation of HLA-DRB1 and HLA-DQB1: (1)The HLA-DRB1 methylation state in Kazakh esophageal carcinoma was lower than in adjacent of cancer and nomal mucosa(P<0.05). The methylation state in Kazakh esophageal carcinoma was higher than in adjacent of cancer and nomal mucosa; The methylation state of HLA-DRB1 was not relationship with age、sex、pathology differentiation、HPV16E6 infection、HLA-II Protein expression and polymorphisms of HLA-DRB1,DQB1 (P>0.05); (2)The HLA-DQB1 methylation state in Kazakh esophageal carcinoma was higher than in adjacent of cancer and nomal mucosa(P<0.05);The methylation state of HLA-DQB1 were not relationship with age、sex、pathology differentiation、HPV16E6 infection、HLA-II Protein expression and polymorphisms of HLA-DRB1,DQB1 (P>0.05); (3) the average rate methylation of HLA-DQB1 CpG6-7, CpG16 -17 in well-differentiated esophageal carcinoma(47.6%和42.6%)was significantly higher than the rate methylation of poorly differentiated carcinoma(18.3%和17.4%)(P <0.05).Conclusion:1. HLA-DRB1*1501 and HLA-DQB1*0301、0602 alleles increase the risk of esophageal carcinoma in Xinjiang Kazakh ethnic ,especially in HPV16E6 infection Kazakh esophageal carcinoma, It may be the factors of genetic susceptibility to Kazakh esophageal carcinoma;2. The methylation of HLA-DRB1 and HLA-DQB1 are related with Kazakh esophageal carcinoma,HLA-DQB1 CpG6-7、CpG16-17 are related with Kazakh esophageal carcinoma pathology differentiation.
