The Downward Regulation of Rats’ OPG Expression in Steroid-induced Femoral Necrosis Rat Models and Its Mechanism

Objective:To induce a steroid-induced femoral necrosis model of rats, observe the expression of OPG and its related cytokines, and discuss the mechanism.Methods:20 Wistar rats were involved in this research, and were randomly divided into two groups, the control group and the glucocorticoid group. The glucocorticoid group was intraperitoneally injected colibacillus endotoxin 20μg/kg per day for two times, then was gluteally injected prednisolone 40ml/kg per day for three times. The control group was injected with the same amount of saline. All the rats were sacrificed in 6 weeks. The content of blood OPG and RANKL were detected by ELISA. Femoral heads of both sides were applied for HE staining, OPG、RANKL andβ—catenin immunohistochemical staining, gene chip analysis and real-time PCR of c-Fos、c-Jun and P-catenin.Results:According to gene chip results, compared to the control group, the glucocorticoid group had a significant low expression of OPG and c-Fos (R>2),while the ratios of c-Jun、RANK、RANKL、PDGF、PTH、PTHR、andβ-catenin were not statistically significant (R <2). The results of ELISA and real-time PCR were similar to that of gene chip. Immunohistochemical staining of OPG andβ—catenin were darker and larger in the control group, the result of RANKL was just the opposite.Conclusion:In the progress of steroid-induced femoral necrosis, glucocorticoids down regulates the expression of OPG, breaks the balance of bone metabolism, and finally leads to osteonecrosis and osteoporosis. In all OPG related cytokines, AP-1 pathway, mainly composed by c-Fos and c-Jun, plays as the main role.
