Abstract

Objective: to study the rabbit bone marrow mesenchymal stem cells and nucleus pulposus cells co-culture between the best proportion of cells. Methods: rabbit bone marrow mesenchymal stem cells and nucleus pulposus cells between respectively with 10:1, 5-1, 3:1, the proportion of 1:1, 1:3, 1:5, 1:10 is divided into 7 groups of mixed culture. 7 consecutive days each co-culture cell growth situation, determined by MTT method to detect each cell proliferation. Toluidine blue stain compare groups of cells proteoglycan synthesis. Immunohistochemical staining to observe Ⅱ compare groups of cells the expression of type collagen. Results: high proportion between bone marrow mesenchymal stem cells in the group than other groups of cell proliferation significantly faster. Between bone marrow mesenchymal stem cells and nucleus pulposus cells with 1:1 and 1:3 proportion co-culture group cell proliferation activity, proteoglycan and collagen type Ⅱ expression is higher than other groups, and between bone marrow mesenchymal stem cells to the nucleus pulposus cell differentiation phenomenon. Conclusion: rabbit bone marrow mesenchymal stem cells and nucleus pulposus cells were cultured cells of the best ratio of 1:1 or 1:3. Between the nucleus pulposus cells and bone marrow mesenchymal stem cells co-culture system in promoting bone marrow mesenchymal stem cells to the nucleus pulposus cells differentiation between.
