Plant-derived proteinase inhibitor genes have been widely used as a main type of resistance genes in insect-resistant transgenic plants. In the present study, the anther-culture lines derived from transgenic rice plants and transgenic rice hybrid F, populations were used as materials. Basta painting and PCR analysis of bar gene and pinll gene and expression of pinll gene in transgenic paternal materials and F2 population CW103 were conducted. The primary results could be summarized as follows:1. Using basta painting and PCR analysis for transgenic paternal materials and part plants in transgenic hybrid F, populations, we found bar gene and pinll gene in rice F; population showed simple 3:1 Mendelian segregation and close linkage. Compared basta painting and PCR analysis of pinll gene, the result indicates basta painting for field rice plants can be used as a convenient and effective method for choosing transgenic rice plants containing pinll gene.2. The survey of inhibitor trypsin activity in CW103,CW103,CW105 showed pinll gene significantly improved the proteinase inhibitor activity in transgenic rice plants. On one hand, the expression of pinll gene displays temporal patterns. With the development of rice growth, the proteinase inhibitor activity in transgenic plants decreases gradually. On the other hand, the expression of pinll gene indicates spatial patterns, showing the highest proteinase inhibitor activity in wounding leaf and the higher activity in leaf up wounding and the lowest in leaf down wounding, which proves the pathway of wounding signal conduction not only conducts upwards but also downwards. In non-transgenic material ZAU11S, however, the change of proteinase inhibitor activity is different with transgenic materials in temporal patterns, though the change in spatial patterns is same.3. Through the proteinase inhibitor activity analysis in’transgenic hybrid F2 population CW103 in heading stage, we found the distribution of the expression of pinll gene is continuous among transgenic plants and can not divided into groups definitely, though pinll gene expressed stably and strongly in parent material and its inherits as a single dominant gene in F2 population. Moreover, the higher expression of pinll gene in hybrid F2than in parent material has been observed.4. The main agronomic traits in three transgenic hybrid F2 populations and one non-transgenic hybrid population have been examined using "bulked segregation" method. The statistics analysis showed transgene results in days to heading elongated, plant height lowed, and fertility decreased. The influence to other agronomic traits is different according to different inheritance background of parents materials. The analysis of variation coefficient of main agronomic traits in F2 population reveals the two transgenes result in much more segregation in rice F2 population, so choose opportunity in F: population will be enlarged.
