Abstract


Before and after culture Objective To compare the K562 cells with the same number as well as a different number of bone marrow mesenchymal stem cells (MSCs) adhesion K562 cell proliferation , cell cycle and apoptosis changes , to study bone marrow mesenchymal stem cells on human leukemia K562 cell growth impact. Determination of the growth curve of K562 cells through the establishment of normal human bone marrow mesenchymal stem cells in vitro culture system and its system of co-culture with K562 cells ; different number of MSCs and K562 cells were co-cultured in the determination of K562 cell proliferation rate curve ; streaming changes of the cytometric detection K562 cell cycle and apoptosis . Results compared to cultured alone K562 cells , co-cultured with the same number of MSCs , K562 cell growth inhibition ; co-cultured with different number of MSCs its antiproliferative effect on K562 cells in a dose - dependent manner ; Cell cycle analysis found K562 cells G0/G1 phase and G2 / M phase of the cell co-culture with MSCs increase and decrease in the S phase of the cell . Co-cultured for 24 h for 48 h , 72 h later by flow cytometry to detect apoptosis rate , found that the rate of apoptosis of K562 cells . Run Conclusion normal human bone marrow mesenchymal stem cells can lead to K562 cell growth inhibition prevents the K562 cell cycle , apoptosis rate within a certain range , with the number of mesenchymal stem cells to increase its anti-proliferative effect on K562 cells enhanced. Objective To observe the bone marrow mesenchymal stem cells (BMMSC) model in rats with acute lymphoblastic leukemia (ALL) leukemia cells ; method will carry a green fluorescent protein (eGFP) gene labeled P388 cells ( mice with acute lymphoblastic leukemia cell lines ) were inoculated intraperitoneally in DBA / 2 mice to establish ALL mouse model intravenous infusion in vitro culture of bone marrow mesenchymal stem cells . Record survival time of mice ; monitoring the mouse blood count ; percentage of eGFP cells in flow cytometry tissue and organ specimens ; observe pathological changes of the various organs of the mice as well as the to detect infusion MSC mice in vivo Y chromosome exist. Results of bone marrow mesenchymal stem cells can extend ALL survival time of the mice , but the blood count changed little ; MSCs can reduce the infiltration of leukemic cells in various organizations and ability to organizations colonization . Conclusion Bone marrow mesenchymal stem cells can affect mice with acute lymphoblastic leukemia (ALL) model the growth of leukemia cells .
