Purpose: long-term physical and mental stress by giving female SD rats, rats POF model to observe the rat estrous cycle, changes in behavioral symptoms, neuroendocrine changes as well as changes in local ovarian factors to study the physical and mental stress-induced mechanism of POF. Methods: SD female rats were randomly divided into normal group and model group. Normal group received no treatment, conventional feeding. Model group, using the modified International Stress day, randomly a given stress, the stress in each cage model rats, the rest of the model rats placed on the side, common in the atmosphere of stress, in order to achieve The purpose of psychological stress. Model set of continuous stress of 28 days, during observation of the estrous cycle of the rat behavior and the last ten days of observation, the normal group and model group were sacrificed on day 29 at the same time, collected rat ovary, serum and brain tissue, HE staining observation ovarian pathological changes of immunohistochemical determination of ovarian of bax and VEGF protein content, serum FSH and E2 content was measured by radioimmunoassay, ELISA method to detect the NE, the corticosterone and INHB content and Realtime-PCR method to detect ovarian GDF-9B mRNA of expressed. Results: behavioral symptoms, compared with the normal group, model group rats fur boring and easy off action to reduce slack in grasping bite reduce wash increased frequency of weight loss. Compared with the normal group, HE staining ovarian pathology change the display model rats at all levels of the normal follicle significantly reduced (P lt; 0.05), a significant increase in atretic follicles (P lt; 0.01), corpus luteum number of significantly reduced (P lt; 0.01 ), and the volume size is uneven; compared with the normal group, model group neuroendocrine changes in serum FSH content increased (P lt; 0.01), E2 decreased (P lt; 0.05); hypothalamic NE value of the model group decreased trend (P gt; 0.05), pituitary NE value reduced (P lt; 0.05), elevated serum NE value (P lt; 0.01); The serum corticosterone significantly higher (P lt; 0.01); the serum INHB significantly decreased (P lt; 0.01); higher than the normal group (P lt; 0.01) compared with the normal group, model group, ovarian bax protein; VEGF protein is lower than the normal group (P lt; 0.01); model of ovarian GDF-9B mRNA expression downward trend, but the difference was not statistically significance (P gt; 0.05). Conclusion: 1. Physical and mental stress can lead to premature ovarian failure, part female, is an important cause of premature ovarian failure. 2. Physical and mental stress can cause the spirit - nerve - endocrine disorder that neurotransmitter secretion disorders, leading to the destruction of the hypothalamic - pituitary - adrenal axis and the hypothalamic - pituitary - ovarian axis imbalance, leading to premature ovarian failure. 3. Physical and mental stress can down-regulate the the female ovarian vascular endothelial growth factor, reducing the occurrence of ovarian blood vessels, ovarian reduced blood supply to change the environment ovary cells increased granulosa cell apoptosis factor expression of bax to influence oocyte GDF-9B mRNA expression The follicular atresia accelerated, eventually jointly lead to premature ovarian failure.
