Abstract


In this study, red raspberry raw ellagic acid extraction, purification and antioxidant activity and antibacterial activity of preliminary research. HPLC determination of ellagic acid in red raspberry investigated the optimum conditions of the solvent extraction of ellagic acid, and its use of high-speed countercurrent chromatography purified the last of the red raspberry ellagic acid oxidation resistance and antibacterial activity. The results show that: ellagic acid as the standard, using high-performance liquid chromatographic method for the determination of ellagic acid in red raspberry. Ellagic acid content in the range of 0.322 ~ 8.05μg a good linear relationship with the peak area, the average recovery of ellagic acid 93.78%, RSD = 0.98%. This method is simple, rapid, accurate, and can be used for the determination of the Red Raspberry ellagic acid content. Solvent extraction of ellagic acid in red raspberry, the main factors affecting the yield orthogonal method to determine the best extraction process. The results show that the various factors on the impact of the order of ellagic acid extraction: acetone concentration gt; feed liquid ratio gt; extraction time gt; extraction temperature, the optimum extraction conditions: 80% acetone solution as extraction solvent, solid to liquid ratio 1:12 (g: mL), extracted under reflux at 80 ℃ 90min. Under this condition, the red raspberry ellagic acid yield was 323μg / g. Using high-speed countercurrent chromatography, to establish a new method of active ingredient in the high-speed countercurrent chromatography separation and purification of mangrove berry ellagic acid. The test results show that: the two-phase solvent system n-butanol: isopropanol: water (v / v / v) = 2:1:3, photogenic as stationary phase, the lower phase as the mobile phase, a flow rate of 2mL/min speed 850r/min, purity 81.6%, 35.2 mg of ellagic acid isolated from the the 120g mangrove berry. Reducing ability was determined using the purified the red raspberry ellagic acid, anti-lard trials, scavenging and anti DPPH oxidation test. The results show that, the Red Raspberry ellagic acid has good reduction capability, can effectively inhibit the oxidation of lard and effective scavenging DPPH · radical, has good antioxidant capacity. Determination of the antibacterial activity of mangrove berry ellagic acid extract common microorganisms. The results show that, the better inhibition of ellagic acid extracts tested bacteria E. coli, Salmonella, Bacillus subtilis. Ellagic acid extract on the test fungi, Aspergillus oryzae, Aspergillus niger, and the test yeasts weak inhibition.
