Abstract


Chinese medicine complex prescription stress the principle of compatibility of monarch,minister, assistant and guide and seven relations.boiling method also follows certain rules, such as decoct first, decoct later, decocting in wrapped condition and so onXu Lingtai Qing Doctors said: "The methods of decoction are important, if the drug is effective , it depends on the methods." It illustrates the importance of preparation of complex prescription.the reason may be the change in amount and qualitative of chemical composition Therefore, the study on the compound constituents with different extraction methods, not only improve clinical medicine, explore and improve the Chinese medicine theory, and promote the Chinese modernization process, but also have the guidance value for the research and development of new drugs. This paper studies on the dynamic changes of chemical composition in Gegenqinlian decoction of different extraction methods. Gegenqinlian decoction is a famous prescription mentioned in shanghanlun written by Zhang Zhongjing. This complex prescription is composed of puerarin, Coptis chinensis .Scutellaria baicalensis and glycyrrhizin,with function of diaphoresis relieving superficies. Modern research shows that the party has functions of fever antibacterial, hypoglycemic, anti-arrhythmic, anti-anoxia, anti-virus, relaxing smooth muscle, and slowing down the heart rate, enhancing immune system and other pharmacological effects. Modern clinical uses it to treat chronic diarrhea, ulcerative colitis, allergic purpura, acute bacillary dysentery, diarrhea in children autumn diseases and diabetes. First this paper established the HPLC fingerprin of Gegenqinlian decoction using the method of high performance liquid chromatography; next compared the fingerprints of single drugs, the negative control solution and the reference standard and the whole side, and assigned 27 peaks on all sides to reflect the material basis for the efficacy of the party. Once again, the analysis of HPLC fingerprint among the combined decoction, and the sub-decoction and the together decoction showed that chemical composition of different types in the Gegenqinlian decoction with different extract has not disappeared, or generate new compounds. Finally the contents of the four major components from the different extracts were determined in the assay, and its content were made analysis of variance by SPSS16.0 software. Consider the content of the major components of the combined decoction as the control group, the contents of puerarin, berberine, baicalin and glycyrrhizin in the sub-decoction increased, the differences on the contents of puerarin, berberine and baicalin between the combined decoction and the sub-decoction were very significant (P <0.01), the difference on the content of glycyrrhizic acid of the sub-decoction was significant (P <0.05). The contents of four index components in the together decoction decreased, the differences on the contents of puerrarin and berberine between the together decoction and the combined decoction were significant (P<0.05),the difference on the contents of baicalin,glycyyhizin were very significant (P <0.01). the differences on the contents of puerarin, berberine and baicalin between the single herb and the combined decoction were very significant (P <0.01), the difference on the content of glycyrrhizic acid of the glycyrrhiz was significant (P <0.05). These results indicate that extraction methods affect the level of contents of the four main indicators in Gegenqinlian decoction. As chemical composition will be different when frying with different methods , which will lead to differences in their efficacy.This article provides a scientific basis for optimization of process and the development of new clinical indications of Gegenqinlian decoction, clearing rationality and integrity of prescription.
