Anti-Quorum Sensing and Anti-Pathogenic Effects of Two Chinese Medicinal Herbs: Galla Chinensis and Coptis Chinensis

Abstract


The exchange of information between quorum sensing (OS) or bacterial bacterial cell density self-coordination reaction is mediated by a hormone-like substance called white-inducer. Gene expression and regulation of the system depends on the OS controls a variety of prokaryotic phenotypes. Taking into account the QS system also controls a variety of harmful bacteria phenotype, including the formation of biofilms, the expression of virulence factors, the motion adhesion, and hence the inhibition the QS is deemed a new method for antimicrobial treatment. This thesis is exploratory research of traditional Chinese medicine plant quorum sensing inhibitors (QSl). The two strains of Chromobacterium violaceum (chromobacterium violaceum) and Agrobacterium the agricultural roots bacteria (Agrobacterium tumefaciens) report bacteria screened from 29 species of plants with anti-QS activity. Of two plants gallnut and Coptis shows inhibitory activity against QS. These two kinds of plants Chromobacterium violaceum a strong QSl activity and in certain dose group suppression Chromobacterium violaceum the CV026 strain produces purple streptozotocin. Tumefaciens agricultural root bacteria A136, Coptis does not affect its QS mediated β-galactosidase expression, its QSl activity range is narrow, and fivefold I have strong activity. Reporter strain Pseudomonas aeruginosa (Pseudomonas aeruginosa) and Escherichia coli (Escherichia coli), and further examination of these plants to QS regulation of virulence factors (groups exercise capacity and biofilm formation of the model strain) influence. The experiment found that a certain dose level, gallic and the Coptis will significantly inhibit P. aeruginosa PAO1 and E. coli populations athletic ability. 100μg/rnL concentration of extract, five times a fiction, respectively, 84%, and 90% of the motility of PAO1 and E. coli groups Coptis respectively 73% and 63% inhibition. The study found that two anti-quorum sensing of Chinese medicine, traditional Chinese medicine has antibacterial activity has targeted QS system, rather than kill bacteria, provide a plan to solve the problem of antibiotic resistance. The results of this study may be active mechanism of traditional Chinese medicine open up a new area, and is expected to provide a new direction of development for the treatment of bacterial infections.
