Abstract


Danshen (Radix et Rhizoma Salviae Miltiorrhiza) lip plants is commonly used bulk herbs in China, with a wide range of pharmacological effects, medicinal usage has increased year by year. Salvia polysaccharide (salvia miltiorrhiza polysaccharideJ, SMP) is a natural polymer, with the regulation of the immune system, the role of the antioxidant and anti-aging, and has a huge potential for development. There are many reports about Salvia study of the chemical composition, but its polysaccharide research, particularly affecting polysaccharide extract the influencing factors less. To this end, the water-soluble fraction extracted from Salvia roots isolated polysaccharide components, explore the influencing factors soluble polysaccharide extraction process, and determine the optimum extraction parameters by orthogonal experiment. Analysis of the monosaccharide composition, and the measured average molecular weight of the polysaccharide. Explore the biological characteristics of the polysaccharide, Salvia polysaccharide antitumor test dose screening and polysaccharides improve the hypoxic tolerance of mice to determine the dose-effect relationship of anti-tumor polysaccharide period Salvia further development and utilization of polysaccharides provide a theoretical basis . The main results are as follows: 1, Salvia water extraction process for Salvia weight with water in the ratio 1:20, extraction time of 4 hours, the ethanol concentration of 90%. Ultrasonic extract the best technology for to Salvia weight and water ratio 1:20, extraction time of 40 minutes, 90% of the ethanol concentration. Both alba or purple flowers Salvia leaves polysaccharide-rich root, followed by the least abundant stem alba root, stem and all parts of the polysaccharide content than alfalfa Salvia various parts. Same period collected different subspecies Salvia polysaccharide content of different content than alfalfa alba Salvia polysaccharide content. The root of the acquisition of the different periods, the polysaccharide content is also different, the root of the polysaccharide content collected in October to May high. 3, the polysaccharide stripped of free protein, obtained through the separation of two homogeneous components, via HPLC analysis of the average molecular weight of the two components were 1.389 x 10 ~ 6,4.028 × 10 ~~ 5. The monosaccharide composition analysis primarily for glucose, galactose, arabinose, rhamnose. 4, the high dose group and the control group compared with the phagocytic rate of macrophages, phagocytic index significant difference (P lt; 0.05), SMP significant increase in mouse peritoneal macrophage phagocytosis rate and phagocytic index or high normal mice nonspecific immune function; cyclophosphamide and high dose group and individual cyclophosphamide group, spleen index, thymus index exists a significant difference (P lt; 0.05), on high doses of Salvia polysaccharide significant improvement cyclophosphamide in the role of immune dysfunction. High dose of the SMP can stimulate T lymphocyte transformation, improving the ability to add value T lymphocytes described Salvia polysaccharide by enhancing immune function to inhibit the growth of tumor cells, and relatively no side effects, and is an ideal anti-tumor medicine. 5 different doses of SMP could degree of extension of the survival time of mice confined under hypoxic conditions, from the survival time of the 100-400ml/kg dose group and the control group were significant differences (P lt; 0.01), which to 200ml/kg extend the maximum rate is better than the other dose groups. 6, by loading swimming duration directly observed Salvia polysaccharide can extend the swimming time in mice, the mechanism of the polysaccharide can improve the body's energy metabolism, reduce protein and nitrogen-containing compounds are decomposed, thereby reducing blood urea nitrogen, anti-fatigue effect.
