Effects of CRZ-1 on Growth of Wheat under Salt Stress, Chlorophyllase and Yield

Abstract


The effects of CRZ-1 on wheat were proved in this paper by cultivate, physiology and enzyme test under salt stress, chlorophyllase (Chlase) activity restraint test, and cultivate in the field. The probable way of effect of CRZ-1 was also studied.Great enhancement effect is showed between 1mg/L-0.01mg/L of concentration of CRZ-1 in the cultivate test under normal growing environment. The curve of concentration-activity is similar as plant growth substances. CRZ-1 may be a new kind of plant growth substances.Effects of CRZ-1 of different concentrations on wheat under 200 mmol·L-1 NaCl salt stress were investigated. The treated groups showed better resistance than CK. High germinative force, greater root length and weight, more chlorophyll content was showed in treated group. Compared with CK, CRZ-1 (10,1,0.1 mg·L-1) could apparently increase the activity by 14.1%,28.7%,15.0% of POD with comparison respectively. 29.2%,175.8%,53.8% of SOD respectively and 13.39%,38.19%,11.81% of CAT respectively, and the same trend showed on electrophoretic band.. Also the content of proline increased, MDA decreased.The test method of activity of chlase was improved in this paper. HPLC method was FIRST used in this test, of which mobile phase was MeOH-HAc (100:0.5). Separation of chlorophyll a (Chl a) was achieved by the method. The restraint of degradation of Chl by Chlase was showed under CRZ-1 treatment in vivo. Restraint rate was 39.89% when concentration of CRZ-1 is 18.75mg/L. It also showed that CRZ-1 could be degradated by Chlase.Two continuous cultivations in the field were taken in this paper. In first year wheat was treated by CRZ-1 under different fertilization, by 0.3mg·L-1 CRZ-1 solution before planting, and 1.6 mg·L-1 four times during the growing period. Measurement of physiological characteristics, yield and quality of wheat showed that, under CRZ-1 treatment, growth of wheat improved, yield increased, and fertilization decreased while quality did not decrease. In the second year wheat was treated by CRZ-1 in different way with full treatment, pre-planting treatment, post-planting treatment and no treatment, while fertilization reduced by half. Two pre-treatment groups were treat by 0.5mg·L-1 CRZ-1 solution before planting, and two post-treatment groups were treated by 1.0 mg·L-1 three times during the growing period. Under CRZ-1 treatment, growth of wheat improved. yield increased as the same as first year. The two tests above were taken in different places and wheat breed, under different temperature and precipitation, and showed great effect when treated by CRZ-1.Totally, great effects of CRZ-1 on wheat were proved in this paper. Higher resistance and greater yield showed when CRZ-1 used. CRZ-1 can also restrain activity of the action of degradation of Chl by Chlase. These results provided foundations to study and appliance on CRZ-1.
