Abstract


Phaneropteridae is an important large family in Tettigonioidea, Orthopera. The species of Phaneropteridae are mainly distributed in tropics and subtropics and are abundant in Oriental Region. It has been recorded more than 30 genera and over 130 species in China. In recent years, development of molecular biological technology promoted great progress of insect systematics and phylogenetics. But reports on phylogeny and molecular evolution of Phaneropteridae are rather few and lack of comprehensive phylogeny. In this research, the phylogenetic relationships in intraspecies, interspecies and intergenus among 10 genera of Chinese Phaneropteridae were discussed based on mitochondrial Cyt b sequence. The results will be helpful to construct a perfect taxonomic system of phaneropteridae and Tettigonioidea.The mitochondrial Cyt b gene of 10 genera and 18 species of Phaneropteridae were sequenced using PCR amplicons directly. Aligned 432bp sequences data were inputted into the software of MEGA2.1 and PHYLIP3.6a for phylogenetic analysis using MP, ML, ME and NJ method with different parameter models, using Tettigonia chinensis and Mecopoda niponensis as outgroups. We aim to clarify the phylogenetic relationships of interfamily, intergenera and inter-species and provide some basic evidences for the further research of taxonomy and phylogeny in Phaneropteridae and Tettigonioidea.The main results of this thesis are as follows:1 Both dryness and stored specimens with ethanol can be used to extract total DNA and amplify target segment of Cyt b gene, PCR production were purified and sequenced directly.2 Cyt b gene of 432bp sequences (delete sequences of the primers) shows strong A/T bias (the average content of A+T is 69.9%), A/T bias is more remarkable at the third position of codon (79.0%), and the variation mainly occurred in the third sites.
