Abstract


Introduction Diabetic nephropathy (Diabetic Nephropathy, DN) diabetes (Diabetes Mellitus, DM) is one of the major microvascular complications , disability and mortality is DM important reason . DM except when hyperglycemia can directly affect the normal metabolism of cells , you can also through non-enzymatic reaction with proteins , lipids and nucleic acids form a large number of advanced glycation end products (Advanced Glycation End-Products, AGEs) , caused by the body structure and function damage. Tissue fibrosis is the most important factor - transforming growth factor (TGF-β) and connective tissue growth factor (CTGF) is upregulated in the DN and its role to play with the AGEs are closely related. Study confirmed that AGEs mediated by the production of reactive oxygen (Reactive Oxygen Species, ROS) is an important intermediate AGEs mediated pathogenic factor . This experiment will study AGEs in cultured renal interstitial fibroblasts (NRK-49F) in the right TGF-β 1 and CTGF mRNA expression and the impact of ROS and antioxidants can inhibit the observation of AGEs induced fibrosis . Materials and methods 1 . Main reagents and instruments AlarmarBlue; DMEM medium ; bovine serum albumin ; FBS ; N- acetylcysteine ​​; 2 ', 7' - dichloro- diacetyl fluorescein ; UNIQ-10 column total RNA was extracted formula purification kit ; TaKaRa RNA PCR Kit (AMV) Ver.3.0. Clean table ; carbon dioxide incubator ; inverted phase contrast microscope ; automatic microplate reader ; flow cytometry ; PTC-100 PCR instrument ; GDS8000 gel automatic imager ; fluorescence spectrophotometer ; speeding cold centrifuge. 2 Experimental Methods 2.1 AGEs-BSA in vitro preparation of AGEs-BSA was prepared according to the literature method in vitro , prepared AGEs-BSA fluorescence spectroscopy
