Abstract


Objective : To study the pigment epithelium -derived factor (Pigment Epithelium Derired Factor, PEDF) for high intraocular pressure -induced ischemia - reperfusion retinal ganglion cells in rats. Method : The rat cornea anterior chamber balanced saline (BSS) infusion to maintain intraocular pressure 110mmHg, in order to prevent the normal retinal blood perfusion . After 60 minutes, remove the infusion needle and restore normal blood flow to the retina , thereby establishing rat retinal ischemia - reperfusion model . Were divided into normal group and non-ischemic retinal ischemia - reperfusion group , which in turn are divided into 0.9% saline injected control group and the experimental group injected PEDF . Reperfusion model to the experimental rats immediately after intravitreal injection 2μg/8μlPEDF, respectively, at 2 and 7 days after injection for enucleation , fixed slices of retinal ganglion cell layer optical microscopy morphology and Fas original in situ hybridization immunological analysis to explore PEDF ischemia - reperfusion retinal ganglion cells in rats. Experimental control group injected with the same method equivalent of 0.9% saline solution. Results: ischemia - reperfusion retinal ganglion cell layer is changed to : cell density reduction , disorganized , there tissue edema , Fas positive cells increased. Reperfusion in two days and seven days, the saline control group and the treatment group PEDF ganglion cells compared with normal control group were significantly reduced (P lt; 0.01) and (P lt; 0.05), saline injection group, the treatment group compared with PEDF ganglion cells to reduce a significant difference (P lt; 0.05), while PEDF injection group ganglion cells were significantly higher than saline injection group (P lt; 0.05); reperfusion two days PEDF treatment group Fas -positive ganglion cells chromatic number and staining was significantly lower than the saline group (P lt; 0.01), two groups of reperfusion 7 days Fas -positive cell counts no significant difference. Conclusion: The retinal ischemia - reperfusion immediately after the line intravitreal injection of PEDF can improve retinal ganglion cell damage ; PEDF may be induced ocular ischemia - reperfusion injury in early retinal ganglion cells have a protective effect.
