Objective: ( a ) studies of pathological liver cell apoptosis and nonalcoholic fatty liver disease relationship . ( 2 ) observation of Cordyceps sinensis and reduced glutathione on liver cell apoptosis and its possible mechanism. Methods: The high fat diet rat nonalcoholic fatty liver disease (NAFLD) models were divided into normal control group , NAFL group ( simple fatty liver group ), NASH group ( non-alcoholic steatohepatitis group ), CS group ( Cordyceps sinensis group ) and GSH group ( glutathione group ) . NAFL start of the experiment the rats were sacrificed at week 8 . 9 weeks in addition to the normal diet group were outside the rest of the rats continued to eat high-fat diet, Cordyceps sinensis group and GSH group while giving the intervention , the experimental animals were sacrificed at 18 weeks . ( 1 ) HE staining of liver tissue slices of liver pathology ; ( 2 ) determination of liver tissue biochemical methods superoxide dismutase (SOD) content ; (3) TUNEL detection of liver hepatocyte apoptosis ; ( 4 ) Immunohistochemistry staining of liver tissue TNF-α, Bax, Bcl-2, Caspase-3, NF-κB P65, TNFR1 protein expression ; (5) ELISA of serum TNF-α levels ; ( 6 ) RT-PCR detection of liver tissue TNF- α mRNA expression levels . Results: ( 1 ) Compared with the normal control group , NAFL group extensive liver steatosis, lipid deposition (P lt; 0.01), hepatocyte apoptosis increased (P lt; 0.01); NASH group to further increase fat deposition in liver cells (P lt; 0.05), accompanied by inflammatory cell infiltration , necrosis and fibrosis, liver cell apoptosis is further increased (P lt; 0.01). ( 2 ) liver SOD levels close to normal and NAFL group (P gt; 0.05), while in the NASH group was significantly lower (P lt; 0.05). ( 3 ) Compared with the normal control group , Bax, Bcl-2, Caspase-3 protein expression in the NAFL group increased (P lt; 0.05), while TNFR1 protein expression did not change significantly (P gt; 0.05), NF-κB P65 showed a non-activated state ; compared with NAFL , Bax, Caspase-3 protein expression (P lt; 0.01),
