Abstract


Research on the yield and quality of rice is undoubted a hot-spot in current agriculture science. It mainly focuses on the content and composition of starch. With the developing of modern molecular biotechnology, starch is studied at molecular level. By analyzing the expression of related genes and the interaction of certain proteins, the pathway and the regulation mechanism of starch synthesis are coming out to us.The formation of ADP-Glucose is a rate-limiting step during starch synthesis. This equilibrium is directly regulated by the supplement of ATP as a substrate, which in turn definite influences the character of plant starch. ATP/ADP translocate protein(AATP), located at the inner membrane of plant plastids, is responsible to the balance of ATP between the stromal and the cytosol. Previous research on transgenic potato indicates that altered AATP activity influences the morphology, yield and starch composition of its tuber.To verify whether, to what extent, the plastidic AATP is contributing to the rate of starch biosynthesis in rice, and its role in regulation of starch pathway, we altered the activity of rice AATP by introducing a corresponding cDNA - increasing it by transforming plant with a sense orientation cDNA of os-aatp(clone from Oryza sativa L.) or ht-aatp(clone from Helianthus tuberosus L. ), and decreasing it by transforming plant with an antisense orientation ones. Further analysis was carried out on them.In this paper, four constructs of either sense or antisense cDNA were introduced into rice Minghui 86(Indica sativa L.) mediated by Agrobacterium.
