Abstract


Objective: To investigate the expression of 5-lipoxygenase. cyclooxygenase-2 and VEGF in colonic carcinoma and it’s relationship with tumour pathological characteristics; To determine whether 5-LOX and COX-2 expression correlate with tumor angiogenesis and identify it’s clinical value in predicting prognosis of colonic carcinoma. Methods: 99 patients with colonic carcinoma were followed up. The expression of 5-lipoxygenase, cyclooxygenase-2 and VEGF were detected by immunohistochemical staining, as well as MVD counting in 99 colonic carcinoma tissue specimens, 14 adenoma tissue specimens and 35 normal colonic mucosal tissue specimens. The relationship of 5-LOX, COX-2, VEGF and MVD with clinical-pathologic factors and prognosis was analysed by SPSS V10.0 for windows software. Results: (1) The survival time of patients was correlated significantly with Duke’s stage , metastasis of colonic carcinoma and treatment with chemotherapy ( P<0.05 ) . ( 2 ) The positive rate of 5-lipoxygenase , cyclooxygenase-2 and VEGF in colonic cancer were significantly higher than those in normal colonic mucosa tissues (P<0.05) .MVD counting in colonic cancer and adenoma tissue was significantly higher than that in normal colonic mucosa tissues ( P<0.01 ) . The expression of 5-lipoxygenase and Cyclooxygenase-2 was correlated significantly with the depth of invasion, the Duke’s stage and the metastasis of colonic cancer (P<0.05) . The positive rate of VEGF and MVD counting were correlated significantly with the Duke’s stage and metastasis of colonic carcinoma (P<0.05) .
