Abstract


The purpose of muscle glycogen synthase (GYS1) is the rate-limiting enzyme gene polymorphism may be associated with type 2 diabetes, insulin resistance , can be used as genetic markers of type 2 diabetes in certain populations of non - oxidative metabolism in skeletal muscle glucose . In this experiment, the Han population living in Tianjin as the research object from unrelated randomly selected 234 cases of type 2 diabetes and 121 normal controls using PCR-RFLP method GYS1 outside 10 M416V exon and intron 14 Xba ⅰ gene polymorphism screening , in order to understand the relationship between these two sites in Tianjin, China gene polymorphism and type 2 diabetes and insulin resistance . Method 1 . Collect blood samples of patients with type 2 diabetes and normal controls . Extract genomic DNA of blood samples . 3 . 234 patients with type 2 diabetes and 121 normal controls using PCR-RFLP method GYS1 outside exon 10 M416V and intron 14 Xba Ⅰ gene polymorphism screening . 4 gene polymorphism screening out sequencing analysis . Analysis of clinical data . Results of GYS1 outside exon 10 M416V loci in 234 cases of type 2 diabetes who were two cases of homozygous mutant genotype , the mutation rate of 0.9% , 53 cases of heterozygous mutant genotype , the mutation rate of 22.6% V allele frequency was 12.2 % ; 121 normal controls detected three cases of homozygous mutant genotype , mutation rate of 2.5% , 25 cases of heterozygous mutant genotypes , mutation rate of 20.7% , V allele frequency of 12.8% between the two groups showed no significant difference ( p gt; 0.05 ) .
