Study on Fermentation and Process of Silage Inoculant Lactic Acid Bacteria

Abstract


Silage inoculant bacteria (SIB), which composed of one or more lactic acid fermenting bacteria and several additives, are used for fermentating green feed materials. The main function of SIB is promoting the microbe ecology, silage process, lactic acid production, and developing qualities of feeds. Lactobacillus Plantarum (LP) and Pediococcus Pentosaceus (PP) are homo-fermentative lactic acid bacteria, with strong ability of lactic acid Priductivity. They act as the former and the latter silage bacteria, respectively.The optimum growth temperature of LP and PP was 37℃ and 3035℃, respectively. In about 12 — 13 hours, both LP and PP entered the logarithmic growth phase. The maxmum biomass of LP was 10×108cfu/g (37℃, 12h) and that of PP was 100×108cfu/g (35℃, 14h). The difference of the optimal growth temperature benefited the silage feed at different silage stages.With agitating and changing the inoculum size, little effect was found on the fermentation. The beginning of logarithmic growth phase was affected by the inoculum size, but little at the end. The fermentation of the LP and PP was promoted most efficientlly by adding yeast extracts, followed by AA-N and NH4-N. The PP absorbed NO3-N, NH4-N, and AA-N. The growth of LP and PP could not be promoted by glucose. The optimum growth pH value of PP and LP was 6.57.0 and 5.56.0, respectively.With the freeze-drying and adding protector method, the bacteria populations of PP and LP was about ten times more than that of the microcapsulation and mid-temperature-drying method. Compared with mid-temperature and microcapsulation method, the actual populations of PP and LP after freeze-drying were 9 times and 15 times more, respectively. In all the experiments, the population of PP was more than that of LP. It was no doubt that freeze-drying benefited PP and LP. The population of PP was 420 ×108 cfu/g by mid-temperature and microcapulation and 4200×108cfu/g byfreeze-drying.Silage inoculant bacteria could be mixed with green feed materials and could be dissolved in water and spraied. Silage inoculant bacteria was composed of some substances benefiting to Silage, including enzyme, glucose ect. The total bacteria of product was 10 X 108cfu/g, the rate of PP and Lpwas 8 and 2. The experimentation shows that the speed of acid producing was increased by the silage inoculant bacteria, which prevented the the second fermentation effectively.
