Abstract

The purpose of all-trans retinoic acid is a hormone-like substances with important physiological functions in vivo physiological concentration is very low. Retinoic acid is also known by the vitamin A (Vitamin A), retinol (retinol) made two steps metabolic, the intermediate product is retinaldehyde. Coenzyme Ⅱ dependent retinol dehydrogenase / visual retinaldehyde reductase (NADP (H)-deperldent retinol dehvdrogerlases / reductases, NRDR) Professor Huang Dongyang 1997 first discovered from rabbit liver and purified and identified important vitamins A metabolic enzyme (Huang and Ichikawa, 1997), is an important member of the short-chain dehydrogenase family, is widely expressed in various tissues of mammals. People with some of the major mammalian NRDR cDNA have been successively cloned. The authors' research group cloned from a human liver an alternatively spliced ??the subtype NRDRiso (AY071856) (Du et al, 2004), suggesting that alternative splicing may play an important role in the functional regulation of the gene in the NRDR and prompts should not be excluded There are other forms of alternative splicing of the gene remains a possibility. Neuroblastoma (neuroblastoma) is the most common pediatric malignant solid tumors, one derived from embryonic neural crest sympathoadrenal system. Retinoic acid caused by axonal formation and neurite outgrowth in vitro induced differentiation of neuroblastoma cell lines. Exogenous retinoic acid can induce neuroblastoma cell lines we have reason to explore the possibility of cells endogenous (physiological) retinoic acid barriers to the normal cell differentiation. Recent data show that abnormal alternative splicing and tumor development related. If the neuroblastoma NRDR2 alternative splicing abnormalities lead to the generation of abnormal proteins, can the normal NRDR reduce the physiological synthesis thus hindering endogenous retinoic acid. Therefore, to study the the neuroblastoma NRDR selective splicing abnormalities may help us to understand the mechanism of development of neuroblastoma. Method using PCR, RACE clones from human neuroblastoma SK-N-SH 23 NRDR alternative splicing isoform which humNRDRA2 green fluorescent protein subcellular localization. The of 23 new NRDR alternative splicing Asian bioinformatics analysis. Analyzed the genetic structure of the NRDR. The results people NRDR genes have three copies, the were DHRS4, DHRS4L2 with DHRS4X. Former two similarity of 97.5%, belonging fragments copy (Segmental Duplication) (90% -98%; ≥ 1 kb). inserted fragments in three copies, respectively life
