Effect of Different Dose Radiation on Nasal Mucosa and Changes of IL-1, IL-6, TNF-Α in the Serum of the Guinea Pig

Abstract


Objective: To investigate different doses of radiation injury in guinea pig nasal, and the use of the ELISA method to detect radiation guinea pig serum IL-1, IL-6, TNF-α content changes. Methods: 125 healthy guinea pigs were randomly divided into a control group and four irradiation group, n = 25, blank control group were sham irradiation, the irradiated group were given the total dose 9Gy 15Gy, 21Gy and 30Gy of nasal radiation, week after irradiation, respectively, in January, February, March, April, the animals were sacrificed 5 each, in the light microscope and electron microscope observation of dynamic pathological changes of the nasal mucosa, the animals were sacrificed before undergoing cardiac puncture blood collection, the serum after centrifugation Detection of IL-1, IL-6, TNF-α content of the change. Results: (1) after irradiation guinea loss of appetite, and hair removal in one week from the skin hyperemia phenomenon, has become more evident with the radiation dose increases. No animal mortality the radiation dose 15Gy below, the animals began to die when the radiation dose up to 21Gy up to 30Gy months when animal mortality rate of 60%. (2) when radiation dose 9Gy, light microscope only see part of the nasal epithelial degeneration, mucosal see a small number of neutrophils and other inflammatory cell infiltration, but yet neatly arranged. Electron microscope, the mild degeneration of the epithelial cells, membrane integrity, prominent cytoplasmic part, the nucleus is still intact, still neatly arranged, cell cilia structure is still normal. Scanning electron microscope, the mucosal epithelium showed no loss, a slight increase of goblet cells, cilia little to reduce still consistent direction. The month began repaired, returned to normal three months. When the radiation dose was 15Gy nasal epithelial degeneration is obvious, partial necrosis, submucosal neutrophils and other inflammatory cell infiltration, goblet fine cells increased secretion enhanced, disordered arrangement of submucosal glands. Electron microscopy of epithelial cell degeneration, of some regions epithelial cells missing, residual epithelial malalignment, cell junction gap widened part of the region cilia disappear, membrane partially dissolved deformed cytoplasmic prominent. Scanning electron microscope, part of the mucosal epithelium, the cilia malalignment, and some lodging, messy, loss of normal morphology, part cilia loss. Began a month to repair, but repair most of the cells lose their normal ciliated columnar epithelial morphology, part of the generation of squamous metaplasia. The radiation dose of 21Gy, nasal epithelial degeneration, malalignment, large areas of necrosis, also reduced basal cell submucosal see a large number of inflammatory cell infiltration, submucosal glands of different sizes, part of the gland is not complete.
