Abstract


Objective: The purpose of this study is to clear cutting FAK ribozyme on the proliferation and apoptosis of gastric cancer cells to FAK experimental material as a target for cancer gene therapy . Methods : Design and Synthesis of for FAK hammerhead ribozyme cDNA sequence , was cloned in the retroviral vector , the ribozyme gene into SGC-7901 cells by liposome -mediated DNA transfection method using RT- PCR, immunocytochemistry detected ribozyme FAK expression in SGC-7901 cells ; ribozyme on proliferation and apoptosis of SGC-7901 cells was observed by MTT assay , TUNEL method . Results: ribozyme transfected SGC-7901 cells ① Immunocytochemistry method detection FAK protein expression in cells significantly inhibited ; ② RT- PCR semi - quantitative detection of FAK mRNA expression levels were significantly decreased . ③ MTT assay cell proliferation activity significantly inhibited ; ④ by TUNEL assay of apoptosis , and apoptosis was 17.5% . Conclusion : ribozyme gene by retroviral vector import SGC-7901 cells can be successfully expressed the the cell FAKmRNA and FAK protein expression levels , and inhibits the proliferation of SGC-7901 cells , while inducing apoptosis .
