Abstract


Objective: To investigate micronized fenofibrate (Liping's) fat-fed spontaneously hypertensive rats (spontaneously hypertensive rats, SHR) Lipotoxicity and intimal hyperplasia, to explore the possible mechanisms for the fibrate clinical applications to provide reliable theoretical basis and experimental evidence. : Caused by balloon injury in rat carotid endarterectomy, 27 SHR were randomly divided into three groups, one group was given standard diet, the other two groups were given high fat diet, high fat diet fenofibrate 100 mg. kg -1 . day -1 gavage. High fat diet calories: 59% fat (fat from lard), 21% protein, 20% carbohydrates. The monthly determination of blood pressure, weight, glucose tolerance was measured every 2 months. Results: (1) compared with the standard diet group, high-fat diet group SHR ??body weight and plasma triglycerides (A55V variant in the UCP2 gene was TG), cholesterol (cholesterol, TC) level was significantly increased, high density lipoprotein cholesterol (high density lipoprotein-cholesterol, HDL-C) levels were significantly lower (P lt; 0.05), rather than fenofibrate treatment group compared with the standard diet group significantly decreased blood pressure, no significant difference in body weight, TG and TC levels, HDL-C was significantly higher. (2) high-fat diet group fasting blood glucose, free fatty acids, glucose curve area was significantly higher than the standard diet group, insulin sensitivity index was significantly lower than the standard diet group (0.0038 ± 0.0007 vs 0.0053 ± 0.0013, P lt; 0.05) Fenofibrate group indicators with the standard diet group was no significant difference (P gt; 0.05) (3) common carotid artery contralateral group W / L was no significant difference (P gt; 0.05); injury side of the high-fat diet group was significantly thicker than the standard diet group (0.36 ± 0.03 vs0.27 ± 0.04, P lt; 0.05 ), local plaque formation, rather than fenofibrate treatment group W / L with standard diet group was no significant difference (P gt; 0.05). (4) compared with the standard diet group, there are a large number of lipid deposition in the high-fat diet group soleus muscle cells
