Objective: To observe the expression of Gaq/11 in lung and spleen of rats during acute lung injury (ALI) and effect of propofol treatment, to explore the pathophysiological mechanism of ALI from molecular level. Methods: 48 Wistar rats were randomly divided into experimental groups (OA groups, n=32) given oleic acid from tail vein (OA 0.2ml/kg), propofol treatment group (P group, n=8) given 1% propofol (80mg/kg/h) from left internal jugular vein for 30 minutes before given the equal dosage OA, and observed for 90 minutes, and control group (C group, n=8) given normal saline (0.2ml/kg). OA groups included 4 groups: OA30min, OA60min, OA90min, and OA120min. Blood gas (pH, PaO2, PaCO2) and MABP indexes, enzyme indexes (SOD, MDA, NO) of plasma and two organs were measured. Gaq/11 concentration in two organs was examined by Western blot. The structure of spleen was observed by both light microscope and transmission electronmicroscope. Results: PaO2 of OA groups and P group reduced (P<0.01), and PaO2 of P group was higher than that of OA 90min group (P<0.01). MABP of OA groups and P group obviously decreased (P<0.05 or 0.01), and MABP of OA 90min group was higher than that of P group (P<0.01).SOD activity in plasma and two organs of OA groups (except OA30min group) and P group was lower than that of C group (P<0.05 或 0.01). SOD activity in plasma and lung of P group was higher than that of OA 90min
