Abstract


Objective: arrhythmia still a morbidity and a high mortality rate of the disease . Actively explore the mechanism of the arrhythmia and seeking safety the efficient antiarrhythmic therapy today . Ibutilide (Ibutilide) is a recently introduced class Ⅲ antiarrhythmic drugs , atrial , ventricular arrhythmias , treatment effects were significant , the most serious clinical applications appear in a small number of patients with drug-induced arrhythmias is torsades de pointes ventricular tachycardia (Tdp) . L- type calcium channel current (ICa-L) is the ion flow , myocardial cell electrical activity and mechanical activity under physiological conditions is mainly involved in the action potential of ventricular muscle fast reaction cell complex formation of polar phase 2 plateau thus affecting the action potential schedule and myocardial effective refractory period . In this study, the Langendorff perfusion apparatus , isolated rabbit single the anterior wall midmyocardium cells (M cells) , were prepared by enzymatic digestion ibutilide a final concentration of 10-5mol / L and 10- 6 mol / L extracellular fluid perfusion cell patch clamp whole-cell recording method to observe the activity of the normal ventricular L-type calcium channel currents (ICa-L) changes from the cellular level explore the ibutilide antiarrhythmic ion mechanism and the emergence of drug-induced heart rate abnormal electrophysiological basis . Materials and Methods Experimental animals : New Zealand purebred rabbits were 40 , provided by the Experimental Animal Center of Hebei Medical University , male or female, weight 2.0 ~~ 2.5kg . 2 cell separation : jarringly rabbit skull occipital stunning rapidly excised cut heart , placed in cold calcium - free Tyrode's solution , cut the pericardium , heart connected to the modified Langendorff heart perfusion apparatus , aortic root retrograde with calcium - free Tyrode solution and 0.4mg/ml collagenase perfusion , accurate separation cut into pieces left ventricular anterior middle
