Abstract


Objective: to establish pig cardiopulmonary bypass heart ischemic preconditioning model , the observed indicators of change in the blood and tissues of myocardial ischemia and reperfusion and ischemic preconditioning , explore cardiopulmonary bypass heart ischemic preconditioning on myocardial cells apoptosis and nuclear factor-κB expression . Methods: 15 healthy test White York pigs ( 4-5 months , weight 36- 40kg ) were randomly divided into three groups . Group 1 pure cardiopulmonary bypass (CPB group , n = 5 ) : do not block the ascending aorta , the parallel loop ; Group 2 ischemic reperfusion group (I / R group , n = 5 ) : parallel circulation 45min after the start of CPB blocking the ascending aorta and inferior vena cava , 90min after opening the ascending aorta and inferior vena cava , parallel loops 120min; group 3 ischemic preconditioning group (IPC group , n = 5) : before clamping of the ascending aorta missing blood pretreatment : blocking the ascending aorta 5min open 10min , repeated three times , the same group . Were taken all the experimental animals before CPB , cardiopulmonary bypass after peripheral venous blood CK and LDH ; aorta after 60min before CPB , before aortic clamping , blocking , take a left ventricular myocardial tissue reperfusion after 90min using immune organization of the SABC method of detection of NF-κB nuclear translocation ; All experimental animals before CPB , respectively, after to take left ventricular myocardial tissue for pathological and transmission electron microscopic examination . After the end of the experiment the left ventricle applications TUNEL assay apoptosis rate . The resulting data (X ± SD) analysis of variance, repeated measurements using SPSS12.0 statistical software results with F value P value indicates P <0.05 considered significant difference . Results: the measured values ??in the three groups of experimental animals before surgery there was no significant difference. (1)
