Abstract


Objective: Asthma is a common pediatric respiratory disease , is characterized by chronic airway inflammation and airway hyperresponsiveness , and clinical often recurrent , serious threat to children's health and growth and development . Its pathogenesis is very complex , found that levels of adenosine in airway inflammatory diseases such as bronchial asthma , bronchospasm and contraction plays an important role . Inhaled adenosine can trigger asthma , bronchospasm . Further studies showed that adenosine role of A2B adenosine receptor ( A2BAR ) on the mast cell membrane , to promote the release of mast cell interleukin- 8 (Interleukin-8, IL-8) and other cytokines , triggering airway inflammation process . Therefore , we speculate the selectivity of A2BAR antagonists can effectively block the airway inflammatory mediators release and bronchospasm . Asthmatic mice of this study was to detect lung tissue A2BAR expression of the optical density of the positive reaction and light gray value bronchial wall thickness of airway smooth muscle , and the level of serum IL-8 , to observe A2BAR antagonist XAC (Xanthine amine congener) mouse asthma model of ovalbumin (Ovalbumin) excited intervention role , to explore A2BAR role in the pathogenesis of asthma airway inflammation , and provide a theoretical basis for clinical application A2BAR antagonist treatment of asthma . Method : Select the mice age six weeks , male , healthy pure BALB / c mice 40 , the adaptive feeding after a week , were randomly divided into three groups : normal control group of 10 , 10 asthma model group , XAC Group 20 ; which XAC group was based on the dose , divided into the XAC small dose group of 10 to 10 the XAC large dose group . Only normal as a normal control group was given saline , are used OVA
