Abstract


Sonic hedgehog (Shh), a vertebrate homolog of the Drosophila segment polarity gene Hedgehog (Hh) one, it encodes a secreted glycoprotein is an important signaling molecule in embryonic development and organ formation play an important role in the process. Bone morphogenetic proteins (Bmps) are members of the TGF-β superfamily signaling molecules play a regulatory function in a variety of developmental processes. Thymus and bursa is a central immune organ, and the bursal is unique birds, they play an important role in the body's immune defense. In the process of the occurrence of a variety of tissues and organs, Shh and Bmp2 jointly participate in the interaction between epithelial cells and stromal cells, and to play a regulatory function in the process of organogenesis. For Shh and Bmp2 in recent years in the thymus of the study focused on the role of T lymphocyte differentiation Shh and Bmp2 pair of human and mouse thymus in the chick embryo thymus, the the bursal development process in Shh and Bmp2 research yet see reported. This test first to use the RT-PCR method the Shh gene 490bp fragment of the chicken, and cloned into pGEM-T easy vector. Using recombinant of Shh-containing plasmid and the existing the Bmp2 gene fragment-containing plasmids were digested linearized DNA was prepared, the preparation of a DIG-labeled in vitro transcription justice and antisense RNA probes. Slices in situ hybridization, Shh and Bmp2 in chick embryo thymus and bursal development process of the temporal and spatial expression pattern. The results show that the thymus during the development of the chick embryo hatch 11 days, Shh expression in the epithelial cells of the epithelial cells and perivascular subcapsular subcapsular, Bmp2 undifferentiated epithelial cells and stromal early differentiation of lymphocytes the expression; hatch 13 days, Shh subcapsular thymus interlobular, expressed in the reticular epithelial cells and stromal cells, Bmp2 lymphocytes in the epithelial cells in the thymus interlobular connective tissue of heterophil granulocytes and thymus intralobular like cells expressed; incubator for 15 days, Shh expression in the thymus interlobular reticular epithelial cells and stromal reticular cells, Bmp2 in the thymus lobules, small lymphocytes and thymus interlobular reticular epithelial cells in the expression; 17 days incubation, Shh save thymic interlobular star reticular epithelial cells and perivascular reticular epithelial cells expression, Bmp2 wrapping the vascular epithelial cells and cortex, small lymphocytes, and medulla
