Abstract


At present , the development of the shrimp farming industry has been well received by the impact of shrimp viruses , shrimp white spot virus ( WhiteSpot Syndrome Virus of WSSV ) disease is one of the most serious diseases in shrimp farming industry research shrimp white spot virus disease has important scientific significance and application value. In this paper, Agaricus blazei (Agaricus blazeiMurill) extract Japanese shrimp (Marsupenaeus japonicus) primary lymphocytes in vitro conditions effect against WSSV infection study designed to find out the of Agaricus extract of WSSV infection antagonistic role lay the foundation for efficient anti - WSSV active substance screening , prevention shrimp white spot virus disease . In order to obtain experimental WSSV samples , the paper first Japanese shrimp by injection WSSV crude extracts infection in vivo , after the PCR detection shrimp really WSSV infection , heart disease shrimp lymphoid tissue and gill separation and preparation of WSSV crude extract . Found that red body , symptoms such as decreased activity of WSSV infected shrimp death; , all seven days the WSSV infection rate of 85 % detected by PCR . Anti WSSV activity in order to extract in vitro studies Agaricus blazei starter Penaeus primary cultured lymphocytes and out the Agaricus blazei the water extract prepared at 4,25 and 60 ° C temperature . WSSV Agaricus blazei water with a concentration of 0.05%, 0.1 % and 0.15% , respectively extracts after 1h infected shrimp primary cultured lymphocytes . The results show that : in vitro WSSV can infect shrimp primary lymphocytes . Agaricus water extract prepared in three different temperatures in different degrees can be suppressed Penaeus lymphocytes WSSV infection cytopathogenic ( cytopathic effect , the CPE ) . PCR detection of WSSV WSSV adsorption and cell infection can not be completely prevented three Agaricus blazei water extracts in vitro . The Agaricus blazei aqueous extract was prepared experimental group 0.1% 25 ℃ and 60 ℃ higher inhibition of WSSV infection compared with other experimental groups . 60 ℃ prepared by different concentrations of alcohol Agaricus blazei aqueous extract further separation of the five kinds of alcohol precipitation samples . Five kinds of products lymphocyte cytotoxicity test in Japanese shrimp found with lymphocytes cultured 2mg/mL alcohol precipitate Japanese shrimp 24h Japanese shrimp cream added to the culture medium
