Study on Pathogenesis of Experimental Staphylococcus Arthritis in Chicken-the Dynamic Changes of Immune Index after Therapy with Levofloxacin

Abstract


To study the mechanism of effect of interleukin-16(IL-1β),interleukin-2(IL-2) and tumor necrosis factor-a(TNF-a) ,and the dynamic changes of other immune index: the phagocytosis of heterophil,the antibody levels and the count of leukocytes, in experimental staphylococcus arthritis from chicken,120 5-week healthy boiler were allotted randomly to 4 groups: the normal control group,the drug control group,the infection control group and the test group, and all immune index were measured in the planned time and their dynamic changes were observed after replicating artificially pathological model with staphylococcus aureus and treating with levofloxacin.Levofloxacin was selected as therapeutic drug considering selecting drug principle in the experiment and the result of drug-susceptibility test.The result about cytokines indicated that IL-1β,IL-2 and TNF-alevels of the infection control group and the test group in serum ,contrasting to the normal control group’s,have wholly increased in defferent degree,and their levels in serum of the test group,comparing with the infection control group’s, had a singnificant difference(P<0.05) at l,7,14,21,28d, at 7,14,21,28,35d and at 5,7,14,28d, respectively ,during the course of the experiment.The result about the sorting count of white blood cell showed that the heterophil counts of the infection control group and the test group within one week, within 3 days, was very significantly higher than the normal control group (p<0.01) respectively,and the heterophil counts of the test group at 5,7,14,21d was significantly lower than the infection control group (p<0.05);the lymphocyte counts of the infection control group at within three weeks, within one week, was very significantly lower than the normal control group (p<0.01) respectively, and the lymphocyte counts of the test group at 5,7,14,21d had a singnificant difference(P<0.05), comparing with the infection control group’s;the mean monocyte counts of the infection control group and the test group peaked 11.5(%)and 12.17(%)at 1d, respectively,and had a very singnificant difference(P<0.01), comparing with the normal control group’s;eosinocyte counts and basocyte counts were ruleless;the drug control group and the normal control group had not singnificant difference.During the course of the experiment,the phagocytose rate of heterophil in the infection control group was significantly higher than the normal group.Moreover, the phagocytose rate of heterophil in the test group gradually rose at 0,1,3,5d,reached a peak at 5d,and gradually decreased to drive normal range ,subsequently. There was not theevident change in the drug control group.Serum conglutinating antibody levels in the infection control group and the test group all sostenuto rose throughout the test. Serum conglutinating antibody levels in the infection control group peaked 1:16 at 35d, and slightly higher than the test group.S.aureus number for peripheral blood in the infection control group and the test group from Od to 3d all rapidly increased.lt was testified by biochemical identify that the biochemical identity of S.aureus isolated from peripheral blood was identical to inoculated S.aureus.The result about Clinical signs showed that injected hocks and opposite hock in the test group and in the infection control group manifested arthrocele at 2d, at 7d respectively. As time passed by, the arthrocele became serious gradually. After chicken in the test group were treated one week, signs the above disappear gradually .However, some chiken in the infection control group walk with a painful tread, even death.And it has been observed from histological section at 35d that histopathological changes of joint in the test group was significantly slighter than the infection control group.There were not evident histopathological changes in the normal control group and the drug control group.Together these results demonstrated that the power contrast of damnification and antidamnification determined direction and final of the inflammatory process, during the course of staphylococcus aureus arthritis from chicken. Bacterial organisms followed by passage of bacteria into the bloodstream and reproduced in sensitive organs or tissues. Lymphocyte % heterophil and monocyte counts relatively rose, and the phagocytose rate of heterophil and serum conglutinating antibody levels rose, which showed that specific and nonspecific immunity reaction all remarkably strengthened. On the other hand, many immune cell which were activated released many inflammatory factor inducing more tissues daminification .After treating with levofloxacin,however,S.aureus content declined ,and decrease the stimulation to host immune system,namely the power of damnification and antidamnification was put down, staphylococcus aureus arthritis from chicken made a gradual recovery.
