Abstract


Ampelopsis (Ampelopsis grossedentata) is Vitaceae Vitis snake in a wild vine , by chemical analysis , rattan tea is rich in flavonoids , a plant , effective parts up to 40 % of the total flavonoids , which dihydrogen myricetin (Dihydromyricetin, referred DMY) content of up to about 27% in recent years, causing great concern . In this paper, rattan tea dihydromyricetin extraction, separation , hypoglycemic and antioxidant activity of a series of studies carried out , the main results are as follows : 1. Dihydromyricetin extraction, separation and structural analysis using ethanol extract of vine tea the total flavonoids, selected by single factor experiment experimental factors and levels, according to two levels of testing to determine the total flavonoids yield on three factors influenced by three factors and three levels of response surface analysis to determine the optimum extraction conditions were: extraction temperature is 70 ℃, with 60 % ethanol , solid-liquid ratio of 1:30 by extraction can make rattan tea flavonoids yield the highest OD values ??obtained with the temperature (x), solvent concentration (y), liquid ratio (z) between the three-factor response surface fitting equation : OD = 1.049833-0.020525x 0.010050y-0.005650z 0.004096x ~ 2-0.000425xy 0.024446y ~ 2 0.020625zx-0.002825zy-0.002854z ~ 2 . Acetone extraction separation of flavonoids dihydromyricetin , obtained by orthogonal experiment the optimum separation conditions for the solid-liquid ratio 1:12, reflux time was 20min, extracted three times. By UV - visible spectroscopy , infrared spectroscopy and HPLC analysis , the sample obtained was treated with acetone is mainly dihydromyricetin . 2 dihydromyricetin hypoglycemic functional studies respectively 10mg/kg.bw, 50mg/kg.bw, 100mg/kg.bw dihydromyricetin dose intraperitoneal injection of alloxan diabetic mice , after the last administration after 10h fasting orbital blood glucose, serum lipids ; thymus , spleen, liver and kidney calculate the relative weight of immune organs ; isolated liver tissue was measured antioxidant index . The results showed that : ① 10mg/kg.bw, 50mg/kg.bw, 100mg/kg.bw dose group could significantly reduce alloxan diabetic mice blood glucose (p lt; 0.01), and high -dose 100mg/kg.bw hypoglycemic effect best . ② dihydromyricetin can make alloxan diabetic mice thymus index and spleen index increase , enhance immunity ; reduced
