Abstract


Objective To study the Guben grams asthma cream containing serum on cultured rat airway smooth muscle cells (airway smooth muscle cells, ASMCs) proliferation and apoptosis and mechanism to explore asthma Su root \ARM) , clarify the reversibility and mechanism of asthma in children , eliminate supplementation and the law provide the basis for anti- asthma relapse in children for clinical use . Containing 10% fetal bovine serum in DMEM SD rat primary cultured ASMCs was examined 4 to 7 cells used in the experiment , MTT (MTT) were applied to observe the the Guben grams gasp cream containing serum on were cultured in vitro the rat of ASMCs growth and IL-4 stimulation of rat ASMCs proliferation model established intervention effect . Light microscope observation of drug-induced morphological changes in the percentage of apoptotic cells were detected by the fragments of DNA in situ end labeling technique (TUNEL) . Results ① 20μL Guben grams asthma cream in doses containing serum to inhibit the growth of ASMCs under normal culture conditions (P lt; 0.05 or P lt ; 0.01 ) . ② Compared with the control group , IL-4 significantly promote ASMCs proliferation ( P lt; 0.01 ) . The high dose of IL-4 the Guben grams asthma cream containing serum IL -4 Guben grams asthma paste dose containing serum , IL-4 DXM -containing serum group , IL-4 INF-γ group than IL- 4 , IL-4 blank serum OD value decreased (P lt; 0.05), but no significant difference between the groups . ③ light microscopy the Guben grams asthma cream in doses containing serum - induced rat ASMCs cell shrinkage , membrane blebbing reduced nuclear staining mass uptake and nuclear rupture . ④ with Guben grams asthma cream high dose treated serum containing apoptotic index was significantly higher than that of the control group and blank serum group (P lt; 0.01 ) . Conclusion Guben grams asthma cream inhibits in vitro cultured rat ASMCs proliferation by inhibiting IL-4 to promote ASMCs proliferation model to induce apoptosis in rat ASMCs , which interfere with the development of asthma ARM , to a certain extent explains asthma reversibility.
