The Detection of Staphylococcus Aureus in Food Using PCR Method

Pathogenic microorganisms of food poisoning caused by contaminated food is a direct result of damage to human health hazards of foodborne poisoning caused by Staphylococcus aureus enterotoxin total bacterial food poisoning in the first place , but there is still a lack of ideal especially rapid method for the detection , so easy to cause the laboratory missed and the delays of the clinical diagnosis . In this study, the method specified by the National Institute of Standards (GB4789.10-2003) and PCR methods of detection of 180 food samples collected from nine areas in Jilin Province , a preliminary understanding of the pollution of the province food in Staphylococcus aureus the two monitoring methods were compared in order to find a more rapid , sensitive , specific , ideal testing methods . The experimental results showed that the PCR method of detecting the positive sample was 45 parts , the positive rate of 25 % , wherein the highest positive rate of fish and cooked food contained within 40%, bags of milk is not detected ; GB detected positive samples is 42 parts , and the positive rate of 23.3% , bulk milk positive rate of up to 37% , bagged milk and vegetables were not detected . Positive PCR assay GB law was no significant difference ( ? 2 = 3.0 , P> 0.05) . PCR method established in this study is designed for the heat-resistant Staphylococcus aureus nuclease gene encoding (nuc gene) a pair of primers , the gene for S. aureus are unique and have a highly conserved , amplification purposes fragment length of 422bp. Create a PCR method in the same time , optimizing the reaction conditions, and the susceptibility test and specificity tests . The test results show that the detection limit of the method 4.32pg of Staphylococcus aureus DNA has good sensitivity ; the test results show that the specificity of detection of the PCR method with Bacillus cereus , Salmonella and other bacteria , Staphylococcus aureus -specific amplification of the target fragment , other bacteria were negative , with good specificity . The PCR method can be completed within 24 hours the GB method takes 4 to 7 days , seen the establishment of Staphylococcus aureus nuc gene PCR method has better sensitivity and specificity , the more quickly and easily than GB .
